Baricitinib, LSN335984 (P‐glycoprotein [P‐gp] inhibitor), and ¹³C‐baricitinib (internal standard) were synthesized in‐house. The ¹⁴C‐baricitinib was obtained from ABC Laboratories (Columbia, MO). The ¹⁴C‐metformin, ³H‐pravastatin, and ³H‐rosuvastatin were purchased from American Radiolabeled Chemicals (St. Louis, MO). The ³H‐vinblastine, ³H‐estrone‐3‐sulfate, ³H‐cholecystokinin fragment 26‐33 amide, ³H‐1‐methyl‐4‐phenylpyridinium, and ¹⁴C‐para‐aminohippuric acid were purchased from Perkin Elmer (Boston, MA). All other chemicals were of analytical grade and purchased from commercial sources.
Stably transfected HEK‐PEAK cells expressing OCT1, OCT2, OATP1B1, OATP1B3, OAT1, OAT3, or vector control (VC), were generated using previously described methods.8 HEK cells transiently transfected with MATE1, MATE2‐K, or VC were purchased from Corning Life Sciences (Transporto cells, Bedford, MA). Madin‐Darby canine kidney (MDCK)‐multidrug resistance protein (MDR)1 cells were obtained from The Netherlands Cancer Institute (Amsterdam, The Netherlands). MDCK‐breast cancer resistance protein (BCRP) cells were generated at Absorption Systems (Exton, PA).

Abemaciclib and its metabolites M2 (LSN2839567) and M20 (LSN3106726) were synthesized by Eli Lilly (Indianapolis, IN). Radiolabeled (¹⁴C) metformin was purchased from American Radiolabeled Chemicals (St. Louis, MO). All other chemicals were of analytical grade and purchased from commercial sources.
