Vitrobot-Enabled Cryo-TEM Sample Prep

A 3 mL droplet of sample solution was placed onto a copper TEM grid covered with a perforated carbon film in a controlled environment vitrifaction system (Vitrobot) at 25 1C and 95% humidity. The excess solution was removed via blotting with filter paper for 3 s. The sample was then placed into liquid ethane prior to storage in liquid nitrogen. The sample was examined with a Tecnai G2 Spirit Twin TEM at 120 kV and the images were captured using a CCD camera (FEI 2K Eagle Camera).
