Cha-GAL80 is a genetic tool used in Drosophila research. It functions as a transcriptional repressor, inhibiting the activity of the GAL4 transcriptional activator in the presence of choline. This allows for spatiotemporal control of gene expression in Drosophila models.

Drosophila Rearing and Genetic Manipulations

Flies (Drosophila melanogaster) were reared on a standard corn meal, yeast and glucose agar medium at 24.5 °C under a 12-h:12-h light: dark (LD) cycle as described before unless otherwise stated10 (link). Control (w1118) and UAS-TrpA1 fly stocks were described previously10 (link),13 (link). Gr5a-GAL4 (#57592), Gr43a-GAL4 (#57636) and Gr64a-GAL4 (#57661) were obtained from the Bloomington Stock Center; Cha-GAL80, Gad-GAL80 and UAS-Shits1 were from Dr. T. Sakai (Tokyo Metropolitan University). To remove possible modifiers and allow for comparisons in a common genetic background, we outcrossed all the strains into a control (w1118) background for at least five consecutive generations.

The following strains were used in this study: fru-GAL4 7 , dsx-GAL4 11 (link) , UAS>stop>TNTin, UAS>stop>TNT 7 , UAS>stop>mCD8::GFP 9 (link) , FLP 335 and FLP 386 were generated as described in previous study 27 (link) , cha-GAL80 39 (link) , tsh-GAL80 from Julie Simpson, and Canton-S strain from the Bloomington Stock Center, Bloomington, IN. Oenocyte less (oe-) ies and their controls were generated as previously described 36 .

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
