Ss carrier dna

Manufactured by Merck Group

SS carrier DNA is a laboratory reagent used in molecular biology and genetics research. It is a single-stranded DNA molecule that serves as a carrier for other DNA samples during various experimental procedures, such as DNA purification or transformation. The primary function of SS carrier DNA is to enhance the efficiency of these processes by providing a substrate for the binding and transport of the target DNA molecules.

Cells were grown and selected for using standard molecular biology techniques (Sherman, 2002 (link)). Yeast transformations were performed using lithium acetate–based methods (Gietz et al., 1995 (link)). In brief, log phase cultures were transformed for 45 min at 42°C with the vector/PCR product in a mixture of ∼40% wt/vol PEG-3350 (Sigma-Aldrich), 0.1 M lithium acetate (Sigma-Aldrich), and 0.28 mg/ml SS carrier DNA (Sigma-Aldrich). Transformants were always selected for on appropriate synthetic media. Table S1 shows the strains used in this study.

Yeast Transformation and Dilution Assays

Cells were grown and selected for using standard molecular biology techniques. Yeast transformations were performed using lithium acetate–based methods (Gietz et al., 1995 (link)). In brief, log phase 5-ml cultures were transformed for 45 min at 42°C with the plasmid/PCR product in a mixture containing ~40% wt/vol PEG-3350 (Sigma-Aldrich), 0.1 M lithium acetate (Sigma-Aldrich), and 0.28 mg/ml SS carrier DNA (Sigma-Aldrich). Dilution assays were spotted on appropriate media by 1:10 serial dilutions and incubated at the indicated temperature.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
