Lgr5-EGFP-IRES-CreERT2 mice (stock number: 008875) and KrasLSL-G12D mice (stock number: 008179) were obtained from Jackson Laboratories. Mex3a KO mice were obtained from Xin Wu's laboratory at Nanjing Medical University. Mex3aflox/flox mice were generated at the Shanghai Model Organisms Center, and first exon (1328 bp) of Mex3a was targeted with flanking LoxP sites, resulting in two LoxP loci.

Conditional Kras Lung Cancer Model

TetO_Lox-Stop-Lox-KrasG12D (tetO_LKrasG12D), p48-Cre, Rosa26-Lsl-rtTA-Ires-Gfp (Rosa_rtTA) and Trp53L strains were donated by Dr. DePinho and described previously27 (link),28 (link). KrasLSLG12D mice were obtained through the Jackson Laboratory. SDCL mice were a generous gift from Dr. Bernfield29 (link). Mice were interbred and maintained on FVB/C57Bl6 hybrid background in pathogen-free conditions at The University of Texas MD Anderson Cancer Center. Animals of both sexes are used in the experiments. Mice were fed with doxycycline water (doxycycline 2g/l, sucrose 20 g/l) where indicated. All manipulations were approved under Institutional Animal Care and Use Committee (IACUC) protocol 00001238. The maximal tumor size allowed by IACUC is 1.5cm in diameter.

Generation of Genetically Engineered Mice

Female mice of the C57BL/6 J strain and Balb/cAnu/nu (nude mice) at 5 weeks of age were purchased from CLEA Japan Inc. (Tokyo, Japan). Conditional knock-in mice heterozygous for the Lox-STOP-Lox-KrasG12D allele (hereafter referred to as KrasLSL-G12D/+ mice) and Trp53flox/flox mice were obtained from the Jackson Laboratory (Bar Harbor, ME, USA) and maintained in a C57BL/6 J background. These mice were intercrossed to generate KrasLSL-G12D/+; Trp53flox/flox mice. Genotyping was performed after weaning as previously described [28 (link),50 (link),]. Animal studies were carried out with the approval of the Chiba Cancer Center for Ethics in Animal Experimentation.

Transgenic Embryonic Mouse Model

The UBC and UCSF Animal Care Committees approved animal experiments. Mice were housed on laboratory chow diet on a 12-hour light-dark cycle. Noon on the morning of the discovery of a vaginal plug was considered embryonic day (E)0.5. CD-1 mice were obtained from Charles River. Frozen embryos of Pdx1-Cre (Gu et al., 2002 (link)) mice were obtained from the MMRRC and were rederived by the BC Preclinical Research Consortium (Vancouver BC, Canada). KrasLSL-G12D mice (Jackson et al., 2001 (link)) and C57BL/6(tetO-Cdkn1b)1Scpr/J mice (Pruitt et al., 2013 ) were purchased from Jackson Laboratory. Sox9-rtTA transgenic mice were made by recombining the reverse tetracycline transactivator (Clontech; pTet-On) into the Sox9 coding region of a 202kb BAC (RP23-36D5); transgenic mice were generated by the UCSF transgenic core as described (Shimajiri et al., 2011 ). Transgene expression was induced in pregnant Sox9-rtTA; tetO-Cdkn1b with daily 1 mg intraperitoneal (IP) injections of doxycycline on indicated gestational days.

Genetically Engineered Mouse Models

Lmo4fl/fl mice were obtained from Prof Visvader (The Walter and Eliza Hall Institute, Australia). Scgb1a1-creER mice were a kind gift from Prof Hogan (Duke University) [30 (link)]. The Lmo4fl/fl and Lmo4LacZ KI/+ mice have been described by Hahm et al. [11 (link)]. All animal experiments were conducted according to the Melbourne Health Research Directorate Animal Ethics Committee guidelines. Mouse tail DNA were genotyped by PCR using the following primers: Lmo4-floxed: 5′-CGAGCTGCTGCCCGGATTCAC-3′, 5′-GCATTCACCAGCCACAGATAAG-3′ and 5′-CGAGCTGAAATTGTCAGCAGCAAG-3′; Shh-cre: 5′-TTCGGCTATACGTAACAGGG-3′, 5′-TCGATGCAACGAGTGATGAG-3′; K-RasLSL-G12D: 5′-CGCAGACTGTAGAGCAGCG-3′, 5′-CCATGGCTTGAGTAAGTCTGC-3′.
