ATR-FTIR Spectroscopic Analysis

Check if the same lab product or an alternative is used in the 5 most similar protocols
To identify the type of material, infrared spectra were acquired using an ATR (attenuated total reflectance)-FTIR spectrometer (FT-IR 4700, Jasco, Japan) equipped with a diamond optical crystal and controlled by Spectra ManagerTM Suite software (Jasco, Japan). FTIR spectra were acquired in the range 4000–650 cm−1, performing a total of 25 scans with a resolution of 4.0 cm−1.

Secondary Structure Analysis of AMS8 Lipase

Check if the same lab product or an alternative is used in the 5 most similar protocols
The secondary structure analysis of the AMS8 lipase was measured using JASCO 815 spectropolarimeter (JASCO, Tokyo, Japan). The secondary structure contents of AMS8 lipase with CaCl2 were valued based on the far-UV (190–260 nm) using a 1 mm path cell length at 20 °C. Refolded AMS8 lipase protein (1 mg/mL) with varying CaCl2 concentrations (0 (control), 20, 40, 60, 80 and 100 mM) was prepared in Tris–HCl (1mM, pH 8.0) buffer. The spectral analysis of AMS8 lipase was subtracted with the subsequent blank containing Tris–HCl (1 mM, pH 8.0). All measurements were repeated three times. The secondary structure contents (beta, helix, turn and random) of AMS8 lipase were analyzed using the method by Yang et al. [38 (link)], using Spectra ManagerTM Suite Software (JASCO, Tokyo, Japan).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
