HEK293T Transfection for Flow Cytometry

HEK293T cells were seeded at 300k cells per well of a 6-well plate. The next day cells were transfected with indicated reporter and any additional plasmids. 24 hour post-transfection cells were trypsinized, spun down at 300g for 5min, and resuspended in DMEM 10% fetal bovine serum. Cells were analyzed on either a BD Bioscience LSR Fortessa or LSR Fortessa X20at the University of California Berkeley flow cytometer facility, and FlowJo. GFP/mCherry ratios were determined from the median GFP and mCherry values. For TPEN and Pomalidomide treatment, TPEN was added at 2.5μM for 16hours and 10μM Pomalidomide (MedChemExpress, HY-10984) for 4 hours.
