Snrnp40^swp/swp mice were crossed with OT-I transgenic or OT-II transgenic mice to produce Snrnp40^swp/swp;OT-I Tg and Snrnp40^swp/swp;OT-II Tg mice (CD45.2). CD4⁺ T cells were isolated from the spleens of Snrnp40^swp/swp;OT-II Tg mice and Snrnp40^+/+;OT-II Tg mice, and CD8⁺ T cells were isolated from the spleens of Snrnp40^swp/swp;OT-I Tg mice and Snrnp40^+/+;OT-I Tg mice, using CD4⁺ T and CD8⁺ T Cell Isolation Kits (Miltenyi Biotec). These cells were then labeled with 3 μM CellTrace™ Far Red. Labeled cells of a single genotype were injected into individual CD45.1 wild-type mice (2 × 10⁶ cells/mouse). The following day, mice were i.p. injected with or without 250 μg of freshly prepared EndoFit™ ovalbumin (OVA, InvivoGen). Two or three days after OVA immunization, spleen cells in recipient mice were harvested for analysis of OVA-specific CD4⁺ T and CD8⁺ T cell proliferation by flow cytometry.

OVA-Specific IgG Response in C57BL/6 Mice

EndoFit™ ovalbumin (OVA) was purchased from Invivogen with
98% purity minimum (SDS-PAGE) and has endotoxin levels <1
EU/mg. Mice (4 per group) were immunized i.m. by 100 µg OVA with
vehicle (propylene glycol) or 5 in vehicle (propylene glycol)
at the indicated doses. After 21 days, serum titers of OVA-specific IgG or
IgG subclasses were measured by ELISA. All experimental procedures using
mice were approved by the Institutional Animal Care and Use Committee
(IACUC) of the University of Texas Southwestern Medical Center, and were
conducted in accordance with institutionally approved protocols and
guidelines for animal care and use. All the mice were maintained at the
University of Texas Southwestern Medical Center in accordance with
institutionally approved protocols.
