Quantifying Gene Expression in Cell Cultures

Hippocampal RNA Quantification and Analysis

Determination of the concentration of total RNA was carried out with the Nanodrop machine and total RNA extraction gained from hippocampal tissues using the Trizol reagent (Sigma, USA). Then, reverse transcription of RNA into cDNA was completed according to the reverse transcription kit instructions (Takara, Japan). The mRNA expression level was detected following the instructions of the fluorescence quantitative kit (Takara, Japan). The 2ΔΔCt method was used for data analysis [21 (link)], and GAPDH served to provide an internal reference. The primer sequences used are displayed in Table 1.
