Immune and Biochemical Markers Evaluation

Blood samples were collected into non-heparinized and K3-EDTA vacuum
tubes (Becton Dickinson Vacutainer Systems) to determine numbers of immune
cells, including leukocytes, lymphocytes, and monocytes, after 3 and 4 weeks.
Immune cell counts were determined using an automatic blood analyzer (ADVIA120;
Bayer, Leverkusen, Germany). ELISA was used to quantify serum concentrations of
tumor necrosis factor (TNF)-α (R&D Systems,
Minneapolis, MN, USA), alanine transaminase (ALT; Sigma-Aldrich, St. Louis, MO,
USA), aspartate aminotransferase (AST; Sigma-Aldrich, St. Louis, MO, USA),
superoxide dismutase (SOD; Cohesion Biosciences, USA), malondialdehyde (MDA;
Abcam, Cambridge, UK), and glutathione peroxidase (GPx; Cayman Chemical, Ann
Arbor, MI, USA).

Recombinant FGF19 and FGF1 Protein Production

Full-length wide type recombinant human FGF19 (FGF19WT) and the FGF1 variant (FGF1ΔHBS) carrying triple mutations (Lys127Asp, Lys128Gln, and Lys133Val) were expressed in E.coli (BL21) and purified as described in the previous studies (Goetz et al., 2007 (link); Huang et al., 2017 (link)). Mouse Total BA Assay Kit was purchased from Crystal Chem INC. Alpha-naphthylisothiocyanate (ANIT) was purchased from Sigma Aldrich (St. Louis, MO, USA). Alanine transaminase (ALT) and aspartate transaminase (AST) Assay Kits were purchased from Sigma Aldrich (St. Louis, MO, USA) and performed in accordance with the protocols provided with manufacturers. All other reagents were of analytical grade.

Antioxidant and Hepatoprotective Rutin Protocol

Quercetin, BHA (butyl-hydroxyanisole) , BHT (butylhydroxytoluene) , TBHQ (tert.butylhydro quinine) , L-ascorbic acid, and linpalmitic acid were purchased from Merck. 1,1-Diphenyl-2-picryl-hydrazyl (DPPH • ) , sodium phosphate buffer, tween 20 and absolute ethanol, Alanine transaminase (ALT) , Aspartate transaminase (AST) , Alkaline phosphates (ALP) , creatinine, and urea kites were purchased from Sigma (Sigma-Aldrich GmbH, Sternheim, Germany) . All other used chemicals were of analytical grade and were obtained from Merck and Sigma (Sigma-Aldrich GmbH, Sternheim, Germany) . Animals were obtained from the animal house -National research centre.
