Ferrocifens Synthesis and Characterization

Materials. All components of conventional blank LNCs are FDA-approved for parenteral administration. Macrogol 15 hydroxystearate (Kolliphor® HS 15) was purchased from BASF (Germany). Phosphatidylcholine from soybean (Lipoïd S 100) was provided by Lipoïd GmbH (Germany) and caprylic/capric triglycerides (Labrafac® WL 1349) were supplied by Gattefosse (France). Ultra-pure water (UPW) was obtained from a Millipore filtration system. All other reagents or solvents used (acetonitrile, methanol, dimethyl sulfoxide DMSO) for chromatography were of analytical grade. Ferrocifens P722, 16 P849, 17 (link) and P769 35 (link) were synthesized as described previously. Esterase from porcine liver was purchased from Merck (SigmaAldrich, Germany). Starting materials for the synthesis of P998 were obtained commercially from Sigma-Aldrich or Alfa Aesar France. Thin layer chromatography was performed on silica gel 60 GF254. IR spectra were obtained on a FT/IR-4100 JASCO 180 spectrometer. 1 H and 13 (link) C NMR spectra were acquired on Bruker 300 or 400 MHz spectrometers. High-resolution mass spectra (HRMS) were acquired in the "Institut Parisien de Chimie Moléculaire (IPCM -UMR 8232)" at Sorbonne Université, Paris.

Xenograft Tumor Growth in Nude Mice

The animal experiments complied with the requirements of the Institutional Animal Care and Use Committee of Shanghai Jiao Tong University (No. 201801054). Four-week-old male nude mice (Lingchang BioTech, n = 6 per group) were injected subcutaneously 5 × 106 HGC27 cells in their front flank. Tumor size was measured every 3 days. The calculation formula of tumor volume is (length × width × height)/2. After 2 weeks, when the size of the engrafted tumor came to about 100 mm3, the mice were injected intraperitoneally either ARV-825 at 10 mg/kg or menstruum control (10% Kolliphor®HS15, BASF) every day. The mice were sacrificed when the tumors in the control group exceeded 1,000 mm3. The tumors were excised and then embedded in paraffin for immunohistochemistry.

Formulation and Characterization of PZQ-loaded Lipid Nanocarriers

All other chemicals were of analytical grade.

Caco-2 Cell Culture and Surfactant Assay

Caco-2 human colonic cancer cells were obtained from the American Type Culture Collection (ATCC; Manassas, Virginia) and were used at passages 35 -50. Cells were cultured in EMEM (Sigma-Aldrich) supplemented with 10% (v/v) FBS (Sigma-Aldrich), 0.1 mg/ml streptomycin, 100 units/ml penicillin, 0.25 µg/ml amphotericin (Sigma-Aldrich) and 2 mM L-glutamine (Sigma-Aldrich) at 37°C with 5% CO2. Unless otherwise stated, cells were seeded at a density of 1 x 10 4 cells per well in 96-well plates (Corning) for 48 hours prior to assay. For the majority of assays the culture media was removed, cells washed twice with phosphate buffered saline (PBS; Sigma-Aldrich) and replaced with Kolliphor® HS15 (BASF) solutions at the required concentrations (0.01, 0.1, 1.0, 10.0, 20.0 or 50.0 mM; concentrations used were selected to be above and below the surfactant CMC 21 ), or the positive control 1.0% (v/v) Triton X-100 (TX; Sigma-Aldrich). All treatment solutions were prepared by direct solvation in Hank's Balanced Salt Solution (HBSS, pH 7.4, Sigma-Aldrich) buffered with 1% HEPES (Biochrom). The negative control in all experiments was 1% HEPES:HBSS solution.

Kolliwax CA, Kolliwax CSA, Kolliwax GMS II, Kolliwax S, Kolliphor P188, Kolliphor ELP, Kolliphor HS15, and Tween 60 were provided by BASF (Ludwigshafen, Germany). Squalene and Squalane were acquired from EFP Biotek (Figueira da Foz, Portugal). Tween 40, Span 20, Span 40, Span 60, and Span 80 were gifted by SEPPIC SA (Paris, France). Water (Ω = 18.2 MΩ.cm, TOC < 1.5 µg/L) was ultrapurified (Sartorius®, Gottingen, Germany) and filtered through a 0.22 µm nylon filter prior to use. All the other reagents were analytical or High Performance Liquid Chromatography (HPLC) grade.

Tacrine was provided from Tocris (Lille, France). Prisma Buffer HT pH 7,4, sink brain buffer, corticosterone and theophylline was obtained from pION (Billerica, Etats-Unis, MA, USA). Methanol, acetonitrile, acetone, water of HPLC grade were purchased from Carlo Erba Reagents (Val de Reuil, France). Formic acid, Na2HPO4·12 H2O, Tris hydrochloride, HEPES and DMSO were obtained from Fisher Scientific (Illkirch, France). NaCl was provided by Acros Organics (Geel, Belgium).
