Western Blot Analysis of CYP3A11

Check if the same lab product or an alternative is used in the 5 most similar protocols
Whole liver extracts were prepared as described previously68 (link), and the protein concentration was determined using the bicinchoninic acid assay according to the manufacturer’s protocol (Pierce, Rockford, IL). Equal amounts of protein (10 mg) were analyzed by SDS-polyacrylamide gel electrophoresis and were transferred onto a nitrocellulose membrane. The membranes were then probed with a rabbit anti-CYP3A11 antibody, followed by probing with a goat anti-rabbit IgG-alkaline phosphatase secondary antibody. The membranes were then washed and incubated with Tropix CDP star nitroblock II ECL reagent as per the manufacturers’ instructions (Applied Biosystems). The membranes were analyzed using a FlourChem FC imaging system (Alpha Innotech). The images were quantified by densitometry using AlphaEase software.

Western Blot Analysis of Mouse Liver Proteins

Check if the same lab product or an alternative is used in the 5 most similar protocols
Mouse liver nuclear and whole cell extracts containing equal amounts of protein were separated by 10% SDS-polyacrylamide gel electrophoresis and transferred to Trans-blot membranes (Bio-Rad, Hercules, CA). Membranes were blocked for 1 h at room temperature in 5% nonfat dry milk or 5% BSA (for phosphoproteins) dissolved in Tris-buffered saline with Tween-20 (TBST) prior to incubation with primary antibodies. Membranes were subsequently washed and probed with a goat anti-rabbit IgG-AP secondary antibody (1:2000) for 1 h at room temperature. The blots were then washed with TBST and incubated with Tropix CDP Star® Nitro block II™ ECL reagent according to the manufacturer’s instructions (Applied Biosystems, Foster City, CA). Protein bands were analyzed and quantified by FluorChem FC Imaging System (Alpha Innotech, San Leandro, CA).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
