Plasma Aβ Levels in TgF344-AD Rats

Blood samples (200–250 μl) were taken from the tail veins of F344 (and TgF344-AD rats. Plasma samples (100 μl) were prepared using 10% potassium K-EDTA mixed with whole blood at final 0.1% K-EDTA dilution. Blood sample were centrifuged at 1500g to extract supernatant plasma. Prepared plasma samples were stored at -80 °C for subsequent biochemical analysis with ELISA. A Meso Scale Discovery (MSD) platform was used to test the samples. Assays for Aβ 40 and 42 were analyzed using V-PLEX Aβ peptide Panel 1, following the manufacturer's instructions (MSD, Gaithersburg, Maryland, USA). An analysis of the variance with Kruskal-Wallis test for plasma concentrations of Aβ42 and Aβ40 in WT F344 and TgF344-AD revealed no significant differences due to sex. Data from males and females was therefore merged prior to analysis for effects of genotype on Aβ42 and Aβ40 (Table S4).

Validating TMT-based Proteomic Profiling

To validate findings from the TMT-based proteomic profiling using an independent method, a Meso Scale Discovery (MSD) platform was used to test the same material for MS analysis. Assays for Aβ, tau, IL-6, and SAA were analyzed using V-PLEX Aβ peptide Panel 1, total Tau kit, Neuroinflammation Panel 1 Human Kit, following the manufacturer’ instructions (MSD, Gaithersburg, MD). Antibodies AT270 and HT-7 (Thermo Fisher Scientific, Waltham, MA) were used to capture/detect pTau181 using the same MSD platform.
