Quantifying Lung Fibrosis in COVID-19

Paraffin-embedded lung sections were dewaxed, rehydrated and stained for 1.5 h with a picrosirius red solution (1.3% picric acid, 1% fast red and 1% fast green). Four or five fields at 4× magnification were taken using a polarized light filter on an Olympus IX71S1F-3 microscope with QCapture Suite Plus (v3.1.3.10) software. Images were quantified (percentage of Sirius red area/total area) using Adobe Photoshop (v 11.0). Pearson correlations between fibrosis score and days from symptom onset to death were calculated for 16 of 19 patients with COVID-19 for whom samples were available and time from symptom onset to death was reported.

Live Imaging of PTU-Treated Embryos

Live imaging was performed on PTU-treated embryos anesthetized in 0.6 mM Tricaine (Sigma, St. Louis, MO, USA, Cat.# A-5040) as per [67 ]. Both live imaging and in situ processed embryos were imaged on a 1% agarose coated dishes using a SZX12 light microscope (Olympus, Tokyo, Japan) with QCapture Suite Plus. White balance and brightness/contrast editing were performed in Adobe® Photoshop® and were performed consistently between all genotypes and conditions.
