Measuring Gastric Tissue MPO Activity

Check if the same lab product or an alternative is used in the 5 most similar protocols
The gastric tissue (100 mg) was homogenized using a small tissue homogeniser in 40 mM phosphate buffer at pH 7.4. The homogenates were centrifuged at 13,000×g for 10 min at 4 °C. MPO activity in the tissue homogenate was estimated as per the protocol described in the commercial MPO kit (BioVision Incorporated, USA). In brief, supernatants were allowed to react with 5, 5-dithiobis-2-nitrobenzoic acid (DTNB) at 25 °C for 120 min and the reaction was terminated by adding the stop mixture. The change in absorbance was measured at 412 nm using a spectrophotometer.

Quantifying Cecal MPO Activity

Check if the same lab product or an alternative is used in the 5 most similar protocols
Myeloperoxidase (MPO) activity in cecal tissue was examined using an MPO kit (BioVision, Milpitas, CA, USA) as per manufacturer’s instructions. Briefly, supernatants from tissue homogenates were mixed with MPO substrate in a 96-well plate and incubated at 25°C for 60min. Reaction was stopped by adding stop mix and quantified using TNB substrate. Absorbance values were read at 412nm using ELISA plate reader (BioTek, Winooski, VT, USA) and enzyme activity was quantified against a calibration curve generated using standards provided in the kit.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
