Neonatal CD4 T Cell Analysis

CBMC of 33 Australian newborns and 39 PNG newborns were studied for CD4 T cell frequencies using flow cytometry. Cells expressing high levels of CD4 were considered to be CD4 T cells, while cells expressing low CD4 levels were considered antigen‐presenting cells (APCs). Briefly, CBMC were resuspended in fluorescence‐activated cell sorting (FACS) buffer (1 × 106 cells/tube), incubated on ice with 50 μl of antibodies for CD4 (BD Biosciences, San Jose, CA, USA) (diluted 1 : 10), washed, and then fixed as per the manufacturer's instructions. Data were acquired using a four‐colour FACSCalibur (BD Biosciences) and results analysed using FlowJo software (Tree Star Inc., Stanford, CA, USA). Dead cells were excluded from analysis using a forward‐ versus side‐scatter gate.
