Transgenic Mouse Models for Cellular Visualization

Keratin19CreER mice (31 (link)) were provided by G. Gu. β-catnflox(Ex3)/+ mice (32 (link)) were bred to K19CreER mice to generate K19CreER;β-catnflox(Ex3)/+ mice. K14-H2BGFP (33 (link)) and Lef1-RFP (7 (link)) transgenic mice were used for visualization of epithelial and mesenchymal cells with the 2-photon microscope. Wntlessfl/fl mice (34 (link)), TcfLef-H2BGFP (35 (link)), Gt(ROSA)26Sortm14(CAG-tdTomato) (tdTomato reporter line) (24 (link)) were obtained from Jackson Laboratories. All procedures involving animal subjects were performed under the approval of the Institutional Animal Care and Use Committee (IACUC) of the Yale School of Medicine.

Lineage Tracing of Cardiac Endothelial Cells

All procedures were approved by the Institutional Animal Care and Use Ethics Committee of Weill Cornell Medical College. Breeding was performed using animals between 12 weeks and 1 year of age. The mouse strains that were intercrossed to generate the animals in this study were: Flk1-GFP (Jackson Labs, 017006), RosatdTom (Jackson Labs, 007905), TCF:LEFH2B-GFP (Jackson Labs, 013752), Cdh5Cre (Jackson Labs, 017968), β-cateninflox/flox (Jackson Labs, 004152), Igs1tm6(CAG-Bgeo,-Wnt5a)Nat (Wnt5aGOF; Jackson Labs, 018141), Wnt5aflox/flox mice (Jackson Labs, 026626), Nkx2.5Cre [provided by Richard Harvey (Stanley et al., 2002)] and β-cateninflEx3/flEx3 [provided by Makoto Taketo (Harada et al., 1999)]. For quantification of NkxEC versus nEC number, 10 µm sections were collected for the entire span of the heart and colocalization masking distinguished EC subtypes.

Lineage Tracing in Developing Mice

All mice used for this study were housed at the Center of Comparative Medicine and Surgery (CCMS), Icahn School of Medicine at Mount Sinai (ISMMS), according to the Institutional Animal Care and Use Committee (IACUC) guidelines (Protocol number LA11-0020). Krt14-Cre, Smoflox/flox, Gli1lacZ, TCF/Lef:H2B-GFP and Rosa26-mT/mG mice were obtained from the Jackson Laboratories. Eedflox/flox mice were kindly provided by Weipeng Mu and Terry Magnuson (Mu et al., 2014 (link)). Ink4a/Arf−/− mice were kindly provided by Philippe Soriano (Serrano et al., 1996 (link)). Both male and female mice were used in this study. Primer sequences used for genotyping are available in Table S4. BrdU was administered to newborn P0 mice by sub-cutaneous injection (50μg per 1g of mouse weight) 4 hours before sacrifice.

Naphthalene-Induced Lung Injury in Mice

Mice were injured with a single IP injection of 300 μg Naphthalene per gram bodyweight and pulsed with nucleotide labels as described in the Supplemental Methods.

Conditional Gfi1b Deletion in Megakaryocytes

Gfi1bfl/fl mice9 (link) were crossed with Rosa-CreERT2, and Axin2+/LacZ (Jackson, B6.129P2-Axin2tm1Wbm/J) or TCF/Lef:H2B-GFP (Jackson, 61Hadj/J) transgene reporter line. PF4-cre mice were used to delete Gfi1b in MKs for immunofluorescence microscopy analysis. Age and sex-matched mice were used in all experiments. For BM transplantation C57B6 CD45.1 mice were used. All mice were housed under SPF conditions and the IRCM Institutional Review Board approved all animal protocols and experimental procedures were performed in compliance with IRCM and CCAC (Canadian Council of Animal Care) guidelines.
