Electrochemical Detection of SARS-CoV-2 Spike Protein

All chemicals were used as received without further purification. The chemicals were purchased as follows: pyrrole 98% − from Alfa Aesar (Germany), sulfuric acid (H2SO4; 96%) from Lachner (Czech Republic), nitric acid (HNO3, 63%), sodium hydroxide (NaOH, 98%), and chloroplatinic acid (H2PtCl6, 40% Pt) from Merck (Germany), potassium phosphate (KH2PO4, 98%) from Honeywell Riedel-de Haen (Germany), sodium chloride (NaCl, 99,5%), potassium chloride (KCl, 99,5%), and disodium hydrogen phosphate (Na2HPO4, 99%) from Carl Roth (Germany). SARS-CoV-2 spike glycoproteins were purchased from UAB Baltymas (Lithuania).
The experiment was performed using potentiostat/galvanostat Metrohm-Autolab model μAutolabIII/FRA2 μ3AUT71079 controlled by NOVA 2.1.3 software (EcoChemie, The Netherlands). All measurements were done in a homemade cell. The total volume of the cell was 250 μL. Three-electrode system consisted of a working electrode (WE) – Pt disk with 1 mm diameter sealed in glass, reference electrode (RE) – Ag/AgCl, and counter electrode (CE) – Pt disk of 2 mm diameter.

Analytical Reagents and Standards

Ultrapure water was obtained by purifying demineralized water in a Direct-Q 3 UV-R system (Merck, Millipore, Darmstadt, Germany). LC-MS-grade acetonitrile, methanol, ammonium formate, and HPLC-grade chloroform were purchased from Carl Roth GmbH and Co. KG (Karlsruhe, Germany). LC-MS-grade isopropanol was from Merck KgaA (Darmstadt, Germany). LC-MS tuning mix and hexamethoxyphosphazine, used as tuning standards, as well as hexakis(1H,1H,3H-tetrafluoropropoxy)phosphazine, and purine, used as lock masses, were purchased from Agilent Technologies (City of Santa Clara, CA, USA). Acetic acid, ethanol, sodium carbonate, disodium hydrogen phosphate, sodium chloride, potassium chloride, and potassium dihydrogen phosphate (all analytical-grade) were from Carl Roth GmbH and Co. KG (Karlsruhe, Germany). 2,2’-Azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS), fluorescein, and trolox (all analytical-grade) were from Sigma-Aldrich Chemie GmbH (Deisenhofen, Germany). 2,2’-Azobis(2-methylpropionamidine) dihydrochloride (AAPH), potassium peroxodisulphate, and gallic acid (both of analytical-grade) were purchased from Fisher Scientific GmbH (Schwerte, Germany). Folin-Ciocalteu phenol reagent (analytical-grade) was obtained from Merck KgaA (Darmstadt, Germany). Acetone (analytical-grade) was obtained from VWR International LLC (Fontenay-sous-Bois, France).

Synthesis and Characterization of Hydrogel Scaffolds

Poly(ethylene glycol) diacrylate (PEG-DA, Mn = 700 g mol−1) and formaldehyde were obtained from Sigma-Aldrich (Merck; Darmstadt, Germany). The radical photoinitiator 1-[4-(2-hydroxyethoxy)phenyl]-2-hydroxy-2-methyl-1-propan-1-one (Irgacure 2959) was a kind gift from Bodo Möller Chemie GmbH (Offenbach, Germany). A Micro BCA Protein Assay Kit was obtained from Thermo Scientific (Waltham, USA). All chemicals and reagent were used as received. Stock solutions containing 28 mg Irgacure 2959 in 3.972 g SPP buffer were obtained by gentle heating with a heat gun at 100 °C until all material was dissolved, and subsequent cooling to room temperature. Water was withdrawn from a Barnstead GenPure xCAD water purification system (Thermo Scientific, Waltham, USA).

Preparation of Bacterial Culture Media

Luria-Bertani broth (LB) and LB agar were purchased from Roth (Karlsruhe, Germany). The 5×M9-minimal salt contained di-sodium hydrogen phosphate (33.9 g/L; Roth, Karlsruhe, Germany), potassium-dihydrogen phosphate (15 g/L; Merck, Darmstadt, Germany), sodium chloride (2.5 g/L; Merck, Darmstadt, Germany) and ammonium-chloride (5 g/L; Merck, Darmstadt, Germany). This M9-salt was diluted 1:5 and supplemented with magnesium sulfate (1 mM, Merck, Darmstadt, Germany), calcium chloride (0.1 mM; Merck, Darmstadt, Germany), glucose (0.4%, Merck, Darmstadt, Germany), thiamin-hydrochloride (10 µg/mL, Roth, Karlsruhe, Germany) and nicotinic acid (0.0025%, Roth, Karlsruhe, Germany). We bought McCoy and RPMI-1640 from Merck (Darmstadt, Germany) and FBS from Thermo Fisher Scientific (Waltham, MA, USA). Human urine was donated by healthy individuals. The urine was filtered (0.2 µm pore size) and used at the day of donation. Potassium chloride came from Merck (Darmstadt, Germany), potassium sulfate from Roth (Karlsruhe, Germany). Phorbol 12-myristate 13-acetat was bought from PromoCell (Heidelberg, Germany). We purchased E. coli LPS K235 from Merck (Darmstadt, Germany), kanamycin, ampicillin and gentamycin from AppliChem (Darmstadt, Germany) and chloramphenicol from Merck (Darmstadt, Germany).
