Manufactured by Merck Group
Sourced in United States, Germany, Sao Tome and Principe, China, Italy, United Kingdom

2′,7′-dichlorodihydrofluorescein diacetate (DCFH-DA) is a fluorogenic compound used as a probe for the detection of reactive oxygen species (ROS) in biological systems. It is a non-fluorescent precursor that can be converted to the highly fluorescent 2′,7′-dichlorofluorescein (DCF) upon oxidation, making it a useful tool for monitoring oxidative stress.

2′,7′-Dichlorodihydrofluorescein diacetate (DCFH-DA; Sigma) was used to measure the ROS levels in the cultured VSMCs, as previously described (18 (link)). Dihydroethidium (DHE; Sigma) staining was used for the in situ detection of ROS levels in frozen tissue sections, which were mounted in OCT-embedding compound and frozen at −20°C. Briefly, unfixed frozen cross-sections were incubated with DHE (5 µmol/l) at 37°C for 30 min in a humidified chamber protected from light, followed by a 5-min wash in PBS. Images were obtained by confocal microscopy (Zeiss LSM 710).

Measuring Oxidative Stress in Bacterial Strains

Check if the same lab product or an alternative is used in the 5 most similar protocols
Triplets for each bacterial strain tested were placed on a 96-well black microtiter plate for cell culture by adding an equal volume of colloidal platinum nanoparticle solution at the appropriate 2 × MIC, 1 × MIC, and ½ × MIC concentrations to the inoculum (0.5 McFarland units). The plate was incubated at 37 °C for 1, 3, 6, 12, and 24 h. After incubation, a peroxynitrite indicator, 2′-7′-dichlorodihydrofluorescein diacetate (DCFH-DA) (Sigma-Aldrich, Gillingham, UK) (final concentration: 5 µM) was added to the wells. In order to achieve better bonding of the dye to ROS, the light-protected plate was placed on an electric mixer for microtiter plates at medium speed (348/1 “ASSISTENT” Rocking shaker, CE type, Karl Hecht GmbH & Co., Sondheim vor der Rhön, Germany). Fluorescence intensity was measured at 485 nm excitation and 525 nm emission (Infinite 200 microplate reader, Tecan Group Ltd., Männedorf, Switzerland) [53 ,54 (link)].

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
