The reagents and chemicals were of analytical grade or higher purity. Additionally, the yeast strain S. cerevisiae STG S101 was purchased from Fermentis (Marcq-En-Baroeul, France).

Quantification of Plasma Kynurenine and Tryptophan

Plasma kynurenine and tryptophan were measured with an Acquity TQ-S UPLC-tandem mass spectrometer (Waters, USA), using reference compounds (L-kynurenine and L-tryptophan) and deuterated tryptophan-d5 and kynurenine-d4 as internal standards (Sigma-Aldrich, South Africa). The plasma samples stored at -70 °C were thawed at room temperature, followed by spiking with internal standards, deproteination with absolute methanol, and vortexing for 10 min. The samples were then centrifuged at 3156 rcf (relative centrifugal force) for 10 min, and 10 μL supernatant was injected in the UPLC-MS/MS system. The analytes were separated using an isocratic elution of injected samples within 2 min. Kynurenine and tryptophan were detected in the multiple reaction mode using electrospray ionization MS (ESI + ) in the positive mode. This method has been previously published ( Adu-Gyamfi et al.,
