Enzymatic Production of Sialic Acid Derivatives

purchased from Research Products International. The protease inhibitor
cocktail, lysozyme, DNase I, UDP-GlcNAc, ManNAc, neuraminic acid,
uridine 5′-diphosphate (UDP), cytidine 5′-triphosphate
(CTP), cytidine 3′,5′-cyclic monophosphate, phosphoenolpyruvate

Vivaspin 20 spin filters and HisTrap and HiTrap Q HP columns were

from Cambridge Isotope Laboratories Inc., and 18O-labeled
water (98%) was obtained from Medical Isotopes Inc.

Neonatal Rat Cardiomyocyte Isolation

Synthesis and Characterization of Ribitol-5-Phosphate

Ribitol (adonitol), D-ribose, ribose-5-phosphate, and cytidine 5’triphosphate (CTP) were purchased from Sigma-Aldrich. Ribitol-5-phosphate was prepared essentially as previously described (Baddiley et al., 1956 (link)). Confirmation of ribitol-5-phosphate was performed by 1D NMR, in addition to observing the complete disappearance of ribose-5-phosphate anomeric signals after reduction by sodium borohydride. Quantification of ribitol-5-phosphate was performed by peak integration of the 1H signals in the 1D 1H NMR spectrum compared to the internal standard 2,2-dimethyl-2-silapentane-5-sulfonate (DSS) at 0.00 ppm (data not shown).
