Breast Cancer Xenograft Model in Mice

Female nude mice (nu/nu Balb/c background, 4–6 weeks old) were purchased from Jackson Laboratories (Bar Harbor, Maine) and cared for in the Animal Core Facility at Case Western Reserve University (Cleveland, OH). All animal experiments were conducted in accordance with an approved protocol by the IACUC of CWRU. Mammary fat pads were injected with 2 x 106 MDA-MB-231-GFP-Luc and 4 × 106 Hs578T-GFP-Luc cells suspended in a matrigel-PBS mixture (1:1). Tumor volumes were monitored weekly using a Vernier caliper. Once the average tumor volume reached 70–90 mm3, mice were subjected to MR imaging (n = 5 for MT218 and n = 5 for Gd(HP-DO3A) (Bracco, Milan, Italy).

Euthanasia and Fixation Protocol for Marine Specimens

Two male and one female adult specimens were collected off the Southern California coast by Aquatic Research Consultants (San Pedro, CA 90732, Fishes4Study.com, Chuck Winkler) and transported to USC the same day. Euthanasia was accomplished by immersion in a 1:3 mixture of isotonic magnesium chloride in artificial sea water. Specimen was maintained in this solution for at least 10 minutes after cessation of breathing, spontaneous behavior and motion of any kind before removing [55 (link), 56 (link)]. Specimen was transferred to 4% paraformaldehyde solution with stirring at 10℃ for several days for fixation. Samples were soaked in 2.5 mM Gadoteridol (Gd-HP-DO3A, Bracco Diagnostics Inc., Princeton, NJ) for 3 days. Samples were immersed in Galden (TMC Industries, MN) while being scanned.

Multiparametric MRI for Prostate Cancer

Routine clinical mpMRI acquisition includes T2W, DCE, and DWI. The DWI includes an apparent diffusion coefficient (ADC) map that was generated at the time of acquisition. Patients were injected with contrast agent Gadoteridol (Gd-HP-DO3A; ProHance, Bracco Diagnostics, Princeton, NJ, USA) with a dose of 0.1 mL/kg before MRI-DCE acquisition. All of the patients were imaged using MAGNETOM-Avanto scanner (Siemens Healthcare, Erlangen, Germany) scanner at 1.5 T with both endorectal coil and phase-array pelvic coil. Table 1 shows more details on the technical parameters of the MRI sequences.
