HPTLC Analysis of Estrogenic Compounds

Both 17β-estradiol (E2, CAS: 50-28-2) and 17α-ethinylestradiol (EE2, CAS: 57-63-6), used as model estrogenic reference compounds, were of the highest analytical grade and were purchased from Sigma-Aldrich. Stock solutions of the reference compounds (5 mg/mL) were prepared in ethanol. Chromatographic separation was performed on silica gel HPTLC plates of type 60G F254 (20 × 10 cm or 10 × 10 cm) purchased from Merck (Darmstadt, Germany). HPTLC solvents were of the highest analytical grade and were purchased from Merck.

Analytical Standards for Environmental Contaminants

Whatman glass fibre filter paper (pore size 0.7 μm) and SPE cartridges Oasis HLB (500 mg, 12 mL) were purchased from Sigma-Aldrich (Aston Manor, South Africa).

Quantitative Analysis of Pharmaceutical Residues

To evaluate the extraction efficiency of these compounds from the HRC tissue and liquid medium, the deuterated labeled compounds sulfamethoxazole-d4 (SMX-d4), trimethoprim-d3 (TMP-d3), and ofloxacin-d3 (OFL-d3) were purchased from Sigma-Aldrich (St Quentin-Fallavier, France) and used as internal standards at a final concentration of 0.1 mg/L. Methanol (MeOH), acetonitrile (ACN), and formic acid (HCOOH) were all HPLC grade (purity > 98.0%) and were purchased from Carlo Erba Reagents S.A.S (Val de Reuil, France). Milli-Q water from a Milli-Q Advantage A10 system (Sigma-Aldrich, St Quentin-Fallavier, France) was used when ultrapure water was required. All individual standard solutions and their stock mixtures were prepared with 100% (v/v) MeOH at a concentration of 1000 mg/L.
