Western Blot Analysis of PD-L1 and Na/K ATPase

Samples were resolved on 4-12% Bis-Tris NuPAGE gels (Invitrogen) according to manufacturer’s recommendations and either stained with Commasie Blue (Bulldog Bio, Portsmouth, NH) or transferred to PVDF membrane for Western Blot staining using iBlot 2 Gel Transfer Device (Invitrogen). Western blot was performed using iBind Automated Western System (Invitrogen) according to manufacturer’s instructions using human anti-PD-L1 antibody (Avelumab, EMD Serona, 1 μg/ml) or rabbit monoclonal Na/K ATPase antibody (Abcam # ab76020, 1:100,000) as primary antibodies and peroxidase-conjugated donkey anti-human IgG-HRP (Jackson ImmunoResearch, 1:2000) or goat anti-rabbit IgG-HRP (Abcam # ab6721, 1:10,000) as sencondary antibodies. Membranes were developed using Clarity Western ECL Substrate (Bio-Rad, Hercules, CA) and images were acquired using Amersham Imager 600 (GE Healthcare) with automated high dynamic range operation.
