Synthesis and Characterization of Silver Nanoparticles

Colistin sulfate and sodium borohydride (NaBH4) were purchased from Sigma Aldrich, Inc., St. Louis, MO, USA. Sodium dodecyl sulfate (SDS), sodium sulfate, sodium phosphate dibasic, and sodium dihydrogen orthophosphate were procured from LOBA Chemie PVT. LTD., Mumbai, India. Silver nitrate (AgNO3), hydrochloric acid (HCl), potassium bromide (KBr), sodium chloride (NaCl), sodium hydroxide (NaOH), dimethyl sulfoxide (DMSO), and acetonitrile (ACN) were analytical grade and procured from RCI Labscan Limited, Bangkok, Thailand. Mueller Hinton (MH) and Luria Bertani (LB) agar and broth were obtained from Titan Biotech Ltd., Delhi, India. Agarose and EDTA were purchased from Bio Basic Canada Inc., Markham, Ontario, Canada. Quick-Loaded® DNA ladder was from New England BioLabs Inc., Herts, England. Safe-Green dye was purchased from Applied Biological Materials Inc., Richmond, BC, Canada. E.Z.N.A.® bacterial DNA kit was obtained from Omega Bio-tek Inc., Norcross, GA, USA. E. coli TISTR 887, P. aeruginosa TISTR 357, and K. pneumonia TISTR 1383 were from the Thailand Institute of Scientific and Technology Research, Pathum Thani, Thailand.

Quantifying MMDA Levels via TBARS

Thiobarbituric acid reactive substance (TBARS) was measured to show MMDA levels in the hippocampus. Tetraethoxypropane (TEP, Sigma Aldrich, Inc., St. Louis, MO, USA) was used as a standard solution. In brief, 100 μL of supernatant from tissue sample was mixed with 100 μL of 8.1% sodium dodecyl sulfate (Loba Chemie, Mumbai, India), 750 μL of 20% acetic acid solution (RCI Labscan, Bangkok, Thailand, pH 3.5), and 750 μL of a 0.8% thiobarbituric acid solution (TBA, Sigma Aldrich, Inc., St. Louis, MO, USA). The specimen was heated in a water bath at 95 °C for 60 min. The mixture was then centrifuged at 4000× g rpm for 10 min. The product of the TBA-MDA reaction was pink and had an absorbance of 540 nm according to spectrophotometric examination.

Lipid Peroxidation Assessment Protocol

MDA levels were measured to analyze lipid peroxidation according to the protocols reported previously [6 (link),21 (link)]. TEP or 1,1,3,3-Tetraethoxypropane (Sigma-Aldrich, St. Louis, MO, USA) was used as a standard solution. The supernatant of the hippocampus or PFC was mixed with 8.1% sodium dodecyl sulfate (Loba Chemie, Mumbai, India), 20% acetic acid (RCI Labscan, Bangkok, Thailand) and 0.8% thiobarbituric acid solutions (Sigma-Aldrich, St. Louis, MO, USA). The mixture was heated at 95 °C in a boiling water bath for 60 min and n-Butanol (RCI Labscan, Bangkok, Thailand) and pyridine (Loba Chemie, Mumbai, India) were added to the mixture and then vortexed. The mixture was centrifuged at 4000 rpm at 25 °C for 10 min. The optical density (OD) was read at 540 nm.

Measurement of Lipid Peroxidation via MDA

The evaluation of MDA was carried out to determine lipid peroxidation. The standard solution was prepared using 1,1,3,3-tetraethoxypropane (Sigma-Aldrich, MO, USA) to produce a calibration line. The supernatant was mixed with 20% acetic acid solutions (RCI Labscan, Bangkok, Thailand), 8.1% sodium dodecyl sulfate (Loba Chemie, Mumbai, India), and 0.8% thiobarbituric acid (Sigma-Aldrich, MO, USA). The reaction mixture was boiled in a water bath at 95°C for 1 h before mixing with pyridine (Loba Chemie, Mumbai, India) and n-butanol (RCI Labscan, Bangkok, Thailand). After centrifugation at 4,000 rpm for 10 min, each sample was evaluated at 540 nm, and the measurement was repeated in triplicate.

Synthesis and Characterization of NR-Based Composites

Natural rubber latex (NR; 60% dry rubber content with high ammonia) was obtained from Chemical and Materials Co., Ltd. (Bangkok, Thailand). Sodium dodecyl sulfate (SDS) and acrylamide (AM) monomer were purchased from Loba Chemie Pvt. Ltd. (Mumbai, India). Acrylic acid (AA) monomer and cumene hydroperoxide (CHP) initiator were obtained from Aldrich (St. Louis, MO, USA). AA was purified by using a column packed with alumina adsorbent before polymerization [24 (link)]. Tetraethylene pentamine (TEPA) activator was purchased from Acros organics (Geel, Belgium). Urea and toluene were purchased from RCI Labscan Limited (Bangkok, Thailand). Terric16A (10 wt%) and 1,3-diphenyl guanidine (DPG) were obtained from the Rubber Authority of Thailand (Bangkok, Thailand). Ethanol was purchased from Duksan Reagents (Ansan, South Korea). The compounding ingredients, such as zinc oxide (ZnO), stearic acid, N-cyclohexyl-2-benzothiazole sulfenamide (CBS), and sulfur, were received from Chemical Innovation Co., Ltd. (Bangkok, Thailand). Precipitated silica (JS-185, surface area (BET) of 170–200 m2/g, a DBP absorption value of 2.0–2.6 cm3/g, and a residue from a 75 µm sieve ≤ 10%) was obtained from Jinsha Precipitated Silica Manufacturing Co., Ltd. (Fujian, China). Silica was used without further purification. Deionized water was used throughout the study.
