Time-lapse imaging of GFP-H2B HeLa cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
Green fluorescent protein-tagged histone H2B-expressing HeLa cells were seeded onto a six-well plate and transfected with siRNA. Twenty-four hours after transfection, two-channel time-lapse video microscopy was performed using a fully motorized Axiovert 200M microscope (Carl Zeiss, Oberkochen, Germany), equipped with AxiocamHRm. Temperature and CO2 control was maintained using the Pecon Incubator (Houston, TX, USA), S-M and heating insert M06 controlled by Temp control (Erbach, Germany), 37-2 and CTI-Controller (Erbach, Germany), 3700. Both phase contrast and green fluorescent protein fluorescence images were acquired for 48 h with a lapse time of 5 min using AxioVision 4.3 software (Oberkochen, Germany). For routine and quantitative analyses, images were acquired using a × 20 objective lens (LD Plan-Neofluar × 20/0.4 Corr Ph2, Carl Zeiss).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
