Aged C57BL/6 Mouse Strain Comparison

The mouse strains used were C57BL/6NTac originally from Taconic (Taconic Biosciences, New York, NY, USA), C57BL/6J distributed by Charles River (Charles River UK), and C57BL/6J mice carrying a BAC transgene to restore murine Nnt (officially C57BL/6J-Tg(RP22-455H18)TG1Rdc/H). All mice were 18-month old males and were no more than 10 generations of mating removed from restock. Animal husbandry, tissue and RNA isolation was performed as detailed in [27 (link)].

Comparison of C57BL/6 Mouse Substrains

All mice were bred, housed, and culled at Medical Research Council Harwell, and therefore, the husbandry was identical for all three substrains. Only male mice were used in this study. Mice were kept and studied in accordance with the UK Home Office legislation and local ethical guidelines issued by the Medical Research Council (Responsibility in the Use of Animals for Medical Research, July 1993; Home Office license 30/3146). Mice were kept under controlled light (light 7 am–7 pm, dark 7 pm–7 am), temperature (21°C ± 2°C), and humidity (55% ± 10%) conditions. They had free access to water (9–13 ppm chlorine) and were fed ad libitum on a commercial diet (SDS Rat and Mouse No. 3 Breeding diet, RM3, 3.6 kcal/g). The mouse strains used were C57BL/6NTac originally from Taconic (Taconic Biosciences), C57BL/6J distributed by Charles River (Charles River UK), and C57BL/6J mice carrying a bacterial artificial chromosome transgene to restore murine Nnt (officially C57BL/6J-Tg(RP22-455H18)TG1Rdc/H). All mice used were no more than 10 generations of mating removed from restock.

Genetically Modified Mice for Immune Research

Murine Cancer Modeling and Evaluation

All animal studies were approved by the NYULMC IACUC (protocol no. IA16-00507). Female B6J mice (C57BL/6J, Taconic), athymic (NCrNU, Taconic) or NSG (The Jackson Laboratory) used in this study were 8–10 weeks old and not involved in previous procedures. All mice were maintained in the animal facility of the NYU Grossman School of Medicine with access to a standard diet (PicoLab Rodent diet 20, catalog no. 5053) and water ad libitum at constant ambient temperature and a 12-h light cycle. Animal numbers were determined according to previous work28 (link),49 (link). Maximum tumor burden was limited to less than 2 cm in any direction; this metric was not exceeded in this study.

Murine Model of B Cell Deficiency

muMT mice, which have a deletion of the IgM heavy chain gene (The Jackson Laboratory, Bar Harbor, ME, USA; JAX stock #002249)(16) and thereby lack mature B cells and immunoglobulins, were backcrossed more than five generations on C57BL/6J background (Taconic Biosciences, Ejby, Denmark), and homozygous muMT and WT littermates were used in the experiments. The following primer pairs were used for genotyping: WT forward primer (5′‐GAA GAG GAC GAT GAA GGT GG‐3′), muMT forward primer (5′‐TTG TGC CCA GTC ATA GCC GAAT‐3′), and a common reverse primer (5′‐CCG TCT AGC TTG AGC TAT TAG G‐3′). The mice were housed at the Sahlgrenska Academy, Laboratory of Experimental Biomedicine in Gothenburg, Sweden, and fed a phytoestrogen‐free pellet diet (Rodent diet Harlan 2016) ad libitum. The study was approved by the animal ethics committee in Gothenburg (1‐2017). Female mice were terminated at different time points: 6 weeks (prepubertal), 12 weeks (postpubertal/young adult), and 16 weeks (adult after the peak growth phase). Male mice were euthanized at 16 weeks of age.

Mlkl Knockout Mouse Alcohol Model

All procedures using animals were approved by the Cleveland Clinic Institutional Animal Care and Use Committee. Mlkl−/− mice were purchased from Taconic Biosciences (#TF2780) and backcrossed to a C57BL/6J background. WT controls were either C57BL/6J purchased from the Jackson Laboratory or WT littermates. WT and Mlkl−/− mice were lethally irradiated at 5 weeks of age and then transplanted with bone marrow from Mlkl−/− or WT mice and allowed to recover for 4 weeks. Mice were then treated with clodronate containing liposomes to deplete resident macrophages in the liver 7 days prior to the start of the Gao‐binge protocol. Mice were allowed free access to a Lieber‐DeCarli liquid diet (Dyets; Cat#710260) containing ethanol at 5% (vol/vol)/28% of total calories or pair‐fed control diet that isocalorically substituted maltose dextrin for ethanol for 10 days. On the final day of the experiment, pair‐fed mice were gavaged with 5 g/kg maltose and ethanol‐fed mice were gavaged with 5 g/kg ethanol in water. Mice were euthanized 6 h after gavage.
