(San Diego, CA). p-SCN-Bn-NOTA (i.e., 2-S-(4-isothiocyanatobenzyl)-1,4,7-triazacyclononane-1,4,7-triacetic acid) was acquired from Macrocyclics, Inc. (Dallas, TX). NHS-fluorescein, hematoxylin staining solution, and Chelex 100 resin (50–100 mesh), tetraethyl orthosilicate (TEOS), triethanolamine (TEA), (3-Aminopropyl)triethoxysilane (APS), Cetyltrimethylammonium chloride solution (CTAC, 25 wt %), copper(II) chloride (CuCl2), sodium sulfide nonahydrate (Na2S·9H2O) and dodium citrate dehydrate were purchased from Sigma-Aldrich (St. Louis, MO). AlexaFluor488- and Cy3-labeled secondary antibodies were purchased from Jackson Immunoresearch Laboratories, Inc. (West Grove, CA). PD-10 columns were purchased from GE Healthcare (Piscataway, NJ). Absolute ethanol, sodium chloride (NaCl), doxorubicin hydrochloride (DOX) were purchased from Fisher Scientific. SCM-PEG5k-Mal was obtained from Creative PEGworks. Water and all buffers were of Millipore grade and pretreated with Chelex 100 resin to ensure that the aqueous solution was free of heavy metals. All chemicals were used as received without further purification.

Radiolabeling of CD31 Antibody with Deferoxamine

ALT-836 was kindly supplied by Altor Bioscience Corp. 1-(4-isothiocyanatophenyl)-3-[6,17-dihydroxy-7,10,18,21-tetraoxo-27-(N-acetylhydroxylamino)- 6,11,17, 22- tetraazaheptaeicosine] thiourea (p-SCN -Bn-Deferoxamine or Df) was purchased form Macrocyclics, Incorporation (Plano, TX) and Chelex 100 resin (50–100 mesh) was acquired from Sigma Aldrich (St. Louis, MO). Primary rat anti-mouse CD31 mAb was purchased for Novus Biologicals (Littleton, CO) and Alexafluor488 and Cy3-labeled secondary antibodies were procured by Jackson ImmunoResearch Laboratories (West Grove, PA). Milli-Q water (resistivity > 18.2 MΩ•cm) was employed in the preparation of all buffers and solutions. Buffers used for radiochemistry were treated with Chelex 100. The rest of the materials and reagents were purchase from Thermo Fisher Scientific Incorporation (Waltham, MA).

AlexaFluor488- and Cy3-labeled secondary antibodies were purchased from Jackson Immunoresearch Laboratories, Inc. (West Grove, CA). PD-10 desalting columns were purchased from GE Healthcare (Piscataway, NJ). 64Cu was produced by a GE PETrace cyclotron using the 64Ni(p,n)64Cu reaction, which has specific activity of > 5 Ci/μmol at the end of bombardment. Water and all buffers were of Millipore grade and pretreated with Chelex 100 resin to ensure that the aqueous solution was free of heavy metals. All other reaction buffers and chemicals were from Thermo Fisher Scientific.

Immunofluorescence Analysis of ALS/FTLD Pathology

Formalin-fixed, paraffin-embedded human hippocampus and cervical spinal cord sections of ALS/FTLD subjects retrieved from the Neurodegenerative Brain Bank at the University of Pittsburgh, following protocols approved by the University of Pittsburgh Committee for Oversight of Research and Clinical Training Involving Decedents (CORID). Following antigen retrieval with Target Retrieval Solution, pH 9 (Dako Agilent), immunofluorescence staining was performed using the following primary antibodies: TIA1 (1:1000, Abeam), Ataxin 2 (1:50, Proteintech), G3BP1 (1:100, Proteintech) and pTDP-43 (1D3, 1:500, kindly provided by Manuela Neumann, Helmholtz Zentrum, Munich, Germany). Immunofluorescence signal was visualized using Alexa Fluor 488 and Cy3 labeled secondary antibodies (both 1:200, Jackson ImmunoResearch) and DAPI nuclear counterstain (1:1000, Thermo Scientific).
