The commercial primary antibodies in this paper were as follows: anti-REPO (8D12, DSHB); anti-pH3 (Ser10) (H0412, Millipore); anti-Cleaved-Caspase 3 (9664, Cell Signaling); anti-α tubulin (ab18251, abcam); anti-MEK (8727, Cell Signaling); anti-p-MEK (Ser217/221) (3958, Cell Signaling); anti-ERK (sc-514302, Santa Cruz); anti-p-ERK (T202/Y264) (4370, Cell Signaling); anti-KCNMA1 (sc-374142, Santa Cruz); anti-Ki-67 (9129, Cell Signaling); and anti-GAPDH (ab8245, abcam).
Three BRAF-related antibodies were used in this study: anti-BRAF (sc-5284, Santa Cruz) were used to detect the total levels of BRAF; anti-BRAFV600E specifically recognizing mutated amino acid sequence (ab228461, abcam) were used to detect the levels of BRAFV600E; anti-p-BRAF (S729) specifically recognizing phosphorylated site (ab124794, abcam) were used to detect the levels of BRAF with serine 729 phosphorylation, to measure the BRAF activation levels regardless of the presence of BRAF mutations.

Molecular Mechanisms of XJR-Induced Apoptosis

Check if the same lab product or an alternative is used in the 5 most similar protocols
After treatment with XJR for 24 h, A549 cells were harvested and lysed on ice in RIPA lysis buffer (Beyotime, Shanghai, China). Protein concentration was detected by using an Enhanced BCA Protein Assay Kit (Beyotime, Shanghai, China). We isolated 40 μg total protein samples by SDS-polyacrylamide gels (PAGE) and electroblotted them onto PVDF-membranes. All membranes were then blocked with 5% defatted milk and then incubated with primary antibodies. Subsequently, membranes were washed with TBST 3 times and incubated with HRP-labeled Goat Anti-Mouse IgG (H+L) antibody (A0216, Beyotime, Shanghai, China). All blots were then developed with an enhanced chemiluminescence (ECL) reagent and analyzed by ImageJ software, which was normalized by GAPDH. Anti-Bcl-2 (4223T), anti-Bax (5023T), anti-cleaved caspase-3 (9661T), anti-cleaved caspase-9 (9509T), anti-MMP2 (40994S), anti-MMP9 (13667T), anti-p38 (8690T), anti-JNK (9258T), anti-p-p38 (4511T), anti-p-ERK (4370T), anti-p-JNK (4668T), and anti-GAPDH (5174S) antibodies were from Cell Signaling Technology (Boston, MA, USA). Anti-ERK (sc-514302) was obtained from Santa Cruz Biotechnology (CA, USA).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
