SRBC and NP-KLH Immunization Protocol

Check if the same lab product or an alternative is used in the 5 most similar protocols
Groups of animals were immunized with SRBC (Innovative Research) or NP-KLH (BioSearch Technologies). For SRBC immunization either a single intra-peritoneal injection of 1×109 SRBC or sequential SRBC injections (Day 0, 1–2×108; Day 5, 1×109) was used as indicated. For NP-KLH,100ug of NP-KLH 1:1 vol/vol in Alum (Immject Alum, Thermofisher) was precipitated and injected intraperitoneally.

Conditional Genetic Manipulation of Immune Cells

Prdm1gfp/+ 11 (link), Prdm1fl 25 (link), Irf4fl 26 (link), Xbp1fl 22 (link) and Rosa26-CreERT2 27 (link) mice were bred and maintained at the animal facilities of the Walter and Eliza Hall Institute. Prdm1fl mice carried a loxP-flanked exon 5, while Prdm1gfp mice expressed an allele truncated after exon 5 (of a total of 7 exons). The PCR analysis for exon 5 loss has been described previously25 (link). All mice were on C57BL/6 background. Animal experiments were conducted according to the protocols approved by the Walter and Eliza Hall Institute ethics committee. Immunization was a single intraperitoneal injection of 100 μg NP-KLH precipitated onto alum (ThermoScientific). For cell transfer experiments, total splenocytes (5 × 106) or isolated B cells (2 × 106) were injected intravenously into Rag1−/− recipients. Cre-mediated deletion of loxP-flanked alleles was triggered by the administration of tamoxifen (Sigma-Aldrich, 0.2 mg/g) by oral gavage on two consecutive days.

NP-Ficoll and NP-KLH Immunization in Mice

Check if the same lab product or an alternative is used in the 5 most similar protocols
NP-Ficoll (conjugation ratio 4:9) and NP-KLH (conjugation ratio 2:3, both from Biosearch Technologies) were dissolved in sterile PBS (Sigma). 25 μg of NP-Ficoll in 200 μl PBS or 100 μg of NP-KLH in 200 μl of PBS/alum adjuvant (mixed at a 1:1 ratio, from Thermo Fisher) was injected into mice intraperitoneally. Blood samples were collected into heparin-coated capillaries by tail vein punctures on indicated days. Plasma was separated by centrifugation and frozen. In certain experiments, the mice were sacrificed 7 days after immunization and tissues were collected for analyses.

Subcutaneous Tumor Modeling and Immunology

Check if the same lab product or an alternative is used in the 5 most similar protocols
The subcutaneous tumor study was performed in NOD/SCIDγ mice (license PPL70/8586), by subcutaneously injecting 107 doxycycline- inducible shRNA- transduced cells with Matrigel (Corning). Tumor volume was calculated by the formula (1/2[length×width2 (link)]). Animals were administered 100 mg/mL doxycycline in drinking water and 0.2 mg/kg intraperitoneally.
Zbp1-/- animals18 (link) were obtained from Manolis Pasparakis, Institute of Genetics (Cologne, Germany). Ten- to 12-week-old littermates were immunized by intraperitoneal injection of 4 mg/kg NP-KLH (Santacruz Biotech) with Alum Adjuvant (Thermoscientific) at a 3:1 ratio followed by a booster dose of NPKLH on day 4. Serum NP-KLH-specific antibodies were measured.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
