Mucosal Immune Markers in Serum and Jejunum

Check if the same lab product or an alternative is used in the 5 most similar protocols
Six males and six females (at 20 wk of age) in each group were selected to collect blood by heart punctures, and then euthanized to collect jejunal mucosal tissue at the end of the experiment. The blood was centrifuged at 4 °C for 10 min at 3000 r/min, and the serum was obtained. The contents of IgA, IgM, and IgG in serum were determined. Jejunal mucosal tissue was taken, and 9 mL normal saline was added to make tissue homogenate. The tissue was centrifuged at 4 °C for 10 min at 3500 r/min, and the supernatant was obtained. The concentrations of secreted immunoglobulin A (SIgA), interleukin-1β (IL-1β), interleukin-8 (IL-8), interleukin-10 (IL-10), interleukin-2 (IL-2), interleukin-6 (IL-6), interleukin-12 (IL-12), tumor necrosis factor-α (TNF-α), and interferon-γ (IFN-γ) were determined. All these indicators were measured using enzyme-linked immunosorbent assay (ELISA) kits (Nanjing Jiancheng Biological Engineering Research Institute, Nanjing, China), and all of them were determined by a full-wavelength micro-plate reader (Tecan Austria GmbH, Groedig, Austria).

MTT Assay for Cell Viability

HuCCT1, QBC939, and TFK1 cells were seeded on 96-well plate at a density of 5,000 cells per well (n=6). After incubation for 48 hrs, 72 hrs, and 96 hrs at 37°C, the cells were incubated with 100 uL MTT (1 mg/mL, Amresco, USA) for 4 hrs. After adding 100 µL of DMSO (Amresco, USA), the value of absorbance was measured at a wavelength of 490 nm using a full-wavelength microplate reader (TECAN, Switzerland).

Cytokine Levels Analysis via ELISA

Check if the same lab product or an alternative is used in the 5 most similar protocols
The levels of IL-2, IL-4, IL-10, and IFN-γ in the serum after allicin intervention were determined using mouse IL-2 (Elabscience, Wuhan, China), IL-4 (Elabscience), IL-10 (Elabscience), and IFN-γ (Elabscience) ELISA kits according to the manufacturer’s instructions. A full-wavelength microplate reader (Tecan, Männedorf, Switzerland) was used to measure the absorbance at a wavelength of 450 nm and calculate the concentration.

Mini-PROTEAN Tetra electrophoresis and blots, a protein semi-dry transfer device, and a gel imaging system were used (Bio-Rad, Hercules, USA). A full wavelength microplate reader (Tecan, Zurich, Switzerland), an automatic high-speed refrigerated centrifuge (Sigma-Aldrich), a SCIENTZ-48 Tissue Grinder (Xinzhi, Hunan, China), an X-Ray Micro-CT SkyScan 1276 (Bruker, Billerica, USA), and a VS200 digital slide scanner (Olympus, Tokyo, Japan) were utilized for tissue analysis.

MTT Assay for PAAD Cell Viability

Check if the same lab product or an alternative is used in the 5 most similar protocols
PAAD cells were inoculated into 96-well plates at 100 μL per well (5×10
3 cells). MTT solution (100 μL, 5mg/mL; Solarbio, Beijing, China) was added at 0 h, 24 h, 48 h and 72 h and incubated for another 4 h. Then the supernatant was discarded and 100 μL DMSO was added to each well to dissolve the formazan crystal. After 10 min, the absorbance of each well was measured at 570 nm with a full-wavelength microplate reader (Tecan, Männedorf, Switzerland), and the cell growth curve was plotted with time as the abscissa and absorbance as the ordinate.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
