PANCREATIC CANCER CELL LINES PROTOCOL

PANC-1, AsPC-1, BxPC-3 and MiaPaca-2 were bought from KeyGEN Biotech (Nanjing, China) and maintained as recommended. The cells cultured in a medium containing different concentrations of DMAPT were grown for 48 h. During this period, all assays, including MTT, cell cycle, Western blotting and IP, were performed every 12 h. All cell lines used in this study were authenticated by short tandem repeat DNA finger-printing and tested for mycoplasma at GENEWIZ, Inc. (Beijing, China) before use. The reports are provided in Supplementary Figure 6.

Pancreatic Cancer Cell Culture

Pancreatic cancer cells PANC-1 and MIA PaCa-2 were purchased from KeyGEN Biotech (China). They were cultured in Dulbecco's modified eagle medium (DMEM) with high glucose from KeyGEN Biotech, supplemented with 10% fetal bovine serum (Biological Industries, Israel), and 100 U ml−1 penicillin and 100 μg ml−1 streptomycin at 37 ℃, 5% CO2. These antibodies were purchased from indicated companies: rabbit anti-RPL10 (Santa Cruz Biotechnology, USA, 1:1000), rabbit anti-β-actin (Proteintech, USA, 1:2000), rabbit anti-OSGIN1 (Abcam, UK, 1:1000), rabbit anti-HMOX1 (Proteintech, USA, 1:1000), rabbit anti-COX4 (Abcam, UK, 1:1000), rabbit anti-LaminB1 (Abcam, UK, 1:1000). Hydrogen peroxide (H2O2) and N-acetylcysteine (NAC) were purchased from Sinopharm Chemical Reagent Company (China) and Beyotime Biotechnology (China) respectively.

Culturing Pancreatic Cancer Cell Lines

The pancreatic cancer cell lines of PANC-1 and Mia PaCa-2 were bought from KeyGEN Biotech Co., Ltd (Jiangsu, China) and cultured with Dulbecco’s modified eagle medium (DMEM, Gibco, California, USA) containing 10% fetal bovine serum (FBS, Biological Industries, Beit-Haemek, Israel) at 37 °C in incubator with 5% CO2. All human cell lines have been authenticated using STR profiling, and mycoplasma contamination was detected to be free. The cells at 80–90% confluence were detached with digestion solution (Gibco, California, USA) and washed with phosphate buffer saline (PBS, Gibco, California, USA).
