Multispectral Imaging for Tumor Analysis

Check if the same lab product or an alternative is used in the 5 most similar protocols
Image acquisition was performed using the Mantra multispectral imaging platform (PerkinElmer, Hopkinton, MA). Areas with non-tumor or residual normal tissue were excluded from the analysis. Representative regions of interest were chosen by the pathologist (S.J.R.), and 3-5 fields of view (FOVs) were acquired at 20x resolution as multispectral images. After image capture, the FOVs were spectrally unmixed and then analyzed using supervised machine learning algorithms within Inform 2.4 (PerkinElmer). Thresholds for positive staining and the accuracy of phenotypic algorithms were optimized and confirmed for each case.

Image acquisition was performed using the Mantra multispectral imaging platform (Vectra 3, PerkinElmer, Hopkinton, MA) as previously described 34 (link). Areas with non-tumor or residual normal tissue (i.e. residual lymph node) were excluded from the analysis. Representative regions of interest were chosen by the pathologist, and 5–7 fields of view (FOVs) were acquired at 20× resolution as multispectral images. Image Analysis was performed using the Inform 2.3 Image Analysis Software (PerkinElmer, Hopkinton, MA).

Multiplex Immunohistochemistry and Multispectral Imaging

Check if the same lab product or an alternative is used in the 5 most similar protocols
Staining was performed using BOND RX fully automated stainers (Leica Biosystems). The target antigens, antibody clones and dilutions for markers, as well as the details of controls are listed in Supplementary Table 6. Image acquisition was performed using the Mantra multispectral imaging platform (Vectra 3, PerkinElmer). Areas with non-tumour or residual normal tissue (that is, residual lymph node) were excluded from the analysis. Representative regions of interest were chosen by the pathologist, and multiple fields of view were acquired at 20× resolution as multispectral images. Cell identification was performed as described39 (link), using supervised machine learning algorithms within Inform 2.3 (PerkinElmer). Thresholds for ‘positive’ staining and the accuracy of phenotypic algorithms were optimized and confirmed by the pathologist (S.J.R.) for each case. The protocol is described in full in the Supplementary Information.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
