Multicolor Imaging of Engineered IRF4-GFP Cells

IRF4-GFP engineered cell lines were plated on a poly-L-lysine coated 15-well μ-Slide Angiogenesis ibiTreat chamber slide (Ibidi) and allowed to adhere for 30 min at 37 °C. Cells were washed with PBS, fixed for 10 min with 4% paraformaldehyde (Electron Microscopy Sciences) and stained with a wheat germ agglutinin Alexa-647 conjugate (5 μg/ml) for 15 min at room temperature. Next, cells were washed twice in PBS and permeabilized in ice cold methanol for 15 min at −20 °C, washed again, and blocked in Duolink Blocking buffer (Sigma) for 1h at room temperature. Cells were stained with conjugated CD34-PE antibody (BioLegend; cat. 343606, RRID:AB_1732008) for 1h at room temperature and imaged on a Zeiss LSM 880 confocal microscope. Images were prepared with FIJI 1.53f51 for display purposes.
