Mast Cell-Deficient Mouse Model

Six-to 8-week-old (young) and 8-to-12 month-old (aged) C57BL/6 wild-type and age-matched mast cell-deficient cKitw-sh mice, congenic to C57BL/6 mice, (Stock No: 012861) were purchased from the Jackson Laboratory (Bar Harbor, ME) for the described experiments. cKitw-sh mice were confirmed for their deficiency in mast cells at the ocular surface and the lacrimal gland using flow cytometry. cKitw-sh mice, unlike the other mast cell-deficient strain (cKitw-v), have a comparable generation of CD45+ immune cells in the bone marrow9 (link),43 (link). The mice were housed in the Schepens Eye Research Institute animal vivarium. All experiments were reviewed and approved by the Schepens Eye Research Institute Animal Care and Use Committee. The mice were treated according to the ARVO Statement for the Use of Animals in Ophthalmic and Vision Research.

Mast Cell Deficiency in Mice

Six- to eight-week-old BALB/c and C57BL/6 wild-type and mast cell-deficient cKitw-sh mice (Stock No: 012861) were purchased from Jackson Laboratory, Bar Harbor, ME for the described experiments. Diverse Mouse and Rat Models for Immunology

Mouse and rat experiments were performed at MIT or the La Jolla Institute for Immunology (LJI). All experimental procedures were approved by institutional IACUC committees. Experiments were done using sex- and age-matched female mice between 6-12 weeks of age. Balb/C mice (strain: 000671) and C57B/6J mice (strain: 000664) were purchased form the Jackson Laboratory. VRC01gHL mice, BCR transgenic knock-in mice carrying inferred germline reverted VRC01 IgH and IgL (34 (link)), were maintained on the B6.GFP (strain 004353; Jackson Laboratory), C57BL/6J or B6.SJL-Ptprcapepcb/BoyJ (B6.CD45.1) (strain 002014; Jackson Laboratory) backgrounds as homozygous lines at LJI. SMARTA mice were maintained on the B6.CD45.1 background at LJI. Mast cell deficient c-Kitw-sh mice (strain 030764; Jackson Laboratory) were used for lymphatic drainage mechanism studies. Female 12-16 wk old Wistar rats (the Jackson Laboratory) were used for imaging antigen drainage from intradermal injections.

Mast Cell-Deficient Mice Model

All animal procedures followed institutional guidelines and were approved by the Institutional Animal Care and Use Committees (IACUC) of Michigan State University (NIH Assurance D16-0054). All procedures followed the recommendations in the ARRIVE guidelines. Male C57Bl/6 mice and male mast cell-deficient (C-kitW-sh) mice (9–12 weeks old; Jackson Laboratory, Bar Harbor, ME USA) were group housed in a temperature- and humidity-controlled environment with a 12-h light/dark cycle and ad libitum access to standard chow and water. Mice were euthanized by intraperitoneal injection of pentobarbital (> 150 mg/kg) followed by decapitation prior to all experimental procedures.

Hematological Profiling of C-Kit w-sh Mice

Animals: Animals were housed in the AAALAC accredited facilities of the CWRU School of Medicine. Husbandry and experimental procedures were approved by the Case Western Reserve University Institutional Animal Care and Use Committee (IACUC) in accordance with approved IACUC protocols 2019-0065. C-Kit w-sh mice were purchased from The Jackson Laboratory. Littermate wild-type control animals were used as comparator animals for all studies and designated "LC", and a combination of male and female mice, aged 8-12 weeks old, were used for all studies.
