ATRP of Functionalized Methacrylates

Ethyl 2-bromo-2-methylpropionate (EBiB, 98%), 2-hydroxyethyl methacrylate (HEMA, ≥99%), oligo(ethylene glycol) methyl ether methacrylate (OEGMA, Mn = 300 g mol−1), copper(i) bromide (CuBr, 98%), benzyl alcohol (BA, ≥99%) and tris[2-(dimethylamino)ethyl]amine (Me6TREN, ≥99%) were purchased from Sigma-Aldrich. HEMA and OEGMA were purified by passing over a short column of activated basic alumina (Sigma-Aldrich) to remove the inhibitor and stored at −27 °C. Copper bromide was purified according to methods described in ref. 49 and 50 (link) and stored under argon at 4 °C. Dimethyl sulfoxide (DMSO, ≥99.5%, for HPLC) purchased from POCH S.A. (Gliwice, Poland) and water (HPLC Plus) purchased from Sigma-Aldrich were purged with argon before use. Concentrated phosphate-buffered saline (PBS, BioUltra) from Sigma-Aldrich was used after tenfold dilution with filtered, deionized water.

Nanomedicine Formulation and Evaluation

All chemical agents including Dextran (DEX, Mw 3500 Da), tris(2-dimethylaminoethyl)-amine (Me6TREN), poly (ethylene glycol) methyl ether methacrylate (OEGMA), hydrazine hydrate and anhydrous solvents were obtained from Sigma-Aldrich (USA). Doxorubicin hydrochloride (DOX•HCl) and other analytical solvent were supplied by Adamas-beta (China). 1,2-distearoyl-sn‑glycero‑3-phosphoethanolamine-N-[methoxy(poly(ethylene glycol))−2000] (DSPE-mPEG2000) was obtained from A.V.T Pharmaceutical Co., Ltd. (Shanghai, China). Biological agents including 3-(4, 5-dimethylthiazol-2-yl)−2,5-diphenyltetrazolium bromide (MTT), Alexa Fluor® 488 phalloidin (AF-488), Lyso-Tracker Green dye and Cy5-NHS were purchased from Life Technologies (USA). TdT-mediated dUTP Nick-End Labeling (TUNEL) apoptosis assay Kit was supplied by Beyotime Biotechnology (China).

Synthesis of Glycinamide-Based Polymers

Glycinamide hydrochloride (98%, Sigma-Aldrich), acryloyl chloride (≥96%, Sigma-Aldrich), potassium carbonate (≥99%, Sigma-Aldrich), 2-chloropropionamide (CPA, 98%, Sigma-Aldrich), copper(i) chloride (CuCl, ≥99%, Sigma-Aldrich) and tris[2-(dimethylamino)ethyl]amine (Me6TREN, 97%, Sigma-Aldrich) were used as received. Ultrapure water was obtained from the Milli-Q water system IQ-7000.
