Cell Culture Conditions for Gastric and Kidney Cell Lines

Gastric cancer cells MKN45 and SGC7901, human embryonic kidney cell line HEK-293T and human gastric mucosa cell line GES-1 were obtained from Shanghai Institutes for Biological Sciences Cell Resource Center. MKN45 and HEK-293T cells were hatched in high-glucose DMEM (Dulbecco’s modified Eagle’s medium) supplemented with 10% FBS (fetal bovine serum, Life Technologies Corporation, Paisley, UK). SGC7901 and GES-1 cells were maintained in RPMI-1640 (Sigma-Aldrich, St. Louis, MO, USA) supplemented with 10% FBS. All cells were incubated in a humidified incubator at 37°C with 5% CO2. The medium was refreshed every 48 h.

Gastric Cancer Cell Line Cultivation

Gastric cancer cell lines MKN-45 and SNU-1 were obtained from Cell Resource Center, Institute of Basic Medicine, Chinese Academy of Medical Sciences. HEK293T cell line was purchased from American Type Culture Collection, Manassas, VA, USA. All cell lines were cultured in RPMI 1640 medium (Meilunbio, Dalian, China) with 10% FBS (Biochannel, Nanjing, China) and 1% penicillin/streptomycin (Beyotime, Beijing, China). The chemical PR-619 (Cat. No. HY-13814) was purchased from MedChemExpress, New Jersey, USA. The primary gastric cancer tissues were received from the First Affiliated Hospital of Soochow University, and the case information was listed in the Supplementary Table 1. The collection and use of human gastric cancer tissues for this study were approved by the Review and Ethics committee of the First Affiliated Hospital of Soochow University.

Acidic Cell Culture Protocol for Cancer Lines

The human cancer cell lines including AGS, HGC27, HeLa, Panc-1, HCT116, A549, and U2OS were purchased from the Cell Resource Center, Institute of Biochemistry and Cell Biology, Shanghai Institutes for Biological Sciences, Chinese Academy of Sciences (Shanghai, China). MKN45 was purchased from the Cell Resource Center, Institute of Basic Medical Sciences, Chinese Academy of Medical Sciences (Beijing, China). Hucct1 were obtained from the Japanese Collection of Research Bioresources (JCRB) (Tokyo, Japan). The cell lines were originally authenticated in China Center for Type Culture Collection (CCTCC) by Short Tandem Repeat (STR) profiling and passaged less than 6 months in the lab. These cells are routinely maintained in the laboratory of our group.The acid cell culture medium (pH 6.5) was obtained by the addition of 1 M HCl solution as previously reported23 (link),24 (link). The pH was estimated by the use of a FE20 FiveEasy Plus pH Meter (METTLER TOLEDO Instruments (China), Shanghai, China).
