Complete Blood Count and Blood Gas Analysis

Check if the same lab product or an alternative is used in the 5 most similar protocols
A complete blood count for each unit was obtained at each timepoint utilizing a Coulter AcT 10 Hematology Analyzer (Beckman Coulter, Brea, CA, USA). An arterial blood gas sample was analysed at each timepoint utilizing VetScan iSTAT (Abaxis, Union City, CA, USA). Prothrombin time evaluation was attempted, but did not generate a result using the iSTAT at any timepoint possibly because of an interaction of the storage solution with the iSTAT cartridge.

Aflatoxin B1 Detoxification via Hemoperfusion

Check if the same lab product or an alternative is used in the 5 most similar protocols
Eighty-one Sprague Dawley rats (350–450 g) cannulated in the jugular and femoral veins (Taconic Labs) were anesthetized with 1.5% isoflurane and injected with 0.5–1.0 mg/kg of AFB1 (1.4 mg/mL solution- 44:56 (v/v) DMF:saline solution) intravenously via the tail vein48 (link)–50 (link). Hemoperfusion was initiated immediately or after a 30, 90, or 240-minute delay and conducted for 4 hours at a flow rate of 1.5 mL/min from the jugular vein through a device containing 3 mL CS polymer beads or an empty device (Control) and back into the rat through the femoral vein catheter. Humane endpoints for euthanasia were included (i.e., inability to eat and drink for 2 days, loss of >15% body weight or >6 °C drop in body temperature). Heparin was dosed at 200 IU/kg/hour during the hemoperfusion and proper heparinization levels monitored by ACT with a Vetscan i-STAT (Abaxis). Rats were observed for a 7-day period following AFB1 exposure with body weight and temperature recorded daily.

Blood Metabolic Profiling in Dairy Cows

Check if the same lab product or an alternative is used in the 5 most similar protocols
Whole blood was sampled by puncture of the jugular vein into 10 mL lithium-heparinized tubes (Vacutainer system, Becton, Dickinson Co.) on d 268 and 271 of gestation from a subset randomly selected blocks that had 28, 32, and 31 cows in the P200, N50, and N150 treatment, respectively, which corresponded to days -6 and -3 relative to calving in the sampled cows. Blood also was sampled from a subset of randomly selected blocks that had 34, 35, and 36 cows in the P200, N50, and N150 treatment, respectively, on d 0, immediately after calving, and at 1, 2, and 4 d postpartum. Whole blood was analyzed within 1 min of sampling using a hand-held biochemical analyzer (VetScan i-STAT, Abaxis). Analytes quantified included pH; blood gases, including partial pressure CO 2 (pCO 2 ), partial pressure of O 2 (pO 2 ), total dissolved CO 2 (TCO 2 ), and saturation of O 2 (sO 2 ); minerals, including iCa, ionized Na (iNa), and ionized K (iK); and concentrations of HCO 3 -, base excess, hematocrit, and hemoglobin.

Venous Blood Collection and Analysis

Jugular venous blood was collected into 10-mL lithium heparin and 10-mL K 2 EDTA evacuated tubes (Vacutainer, Becton Dickinson, Franklin Lakes, NJ) on the day preceding the IVGTT. Samples of whole blood from the lithium heparin tubes were analyzed within 2 min of collection for pH, concentration of HCO 3 -, base excess, partial pressure of CO 2 , total dissolved CO 2 , partial pressure of O 2 , and oxygen saturation using a hand-held biochemical analyzer (VetScan i-STAT, Abaxis, Union City, CA).

Plasma Mineral Profiles in Newborn Calves

Check if the same lab product or an alternative is used in the 5 most similar protocols
Blood samples were collected via venipuncture of the jugular vein into 10-mL tubes containing sodium heparin (Vacutainer, Becton Dickinson, Franklin Lakes, NJ) at birth, before colostrum feeding, and at 1 (24 ± 3 h), 2, 3, 21, and 42 d of age. Within 20 min of collection, plasma was separated by centrifugation at 200 × g for ), Na, K, and ionized Ca (iCa) concentrations using a handheld biochemical analyzer (CG8+; VetScan iSTAT, Abaxis, Union City, CA). Plasma samples were analyzed for total Ca (tCa) and Mg concentrations using an atomic absorption spectrophotometer (AAnalyst 200, Perkin-Elmer Inc., Waltham, MA) according to procedures previously described by Martinez et al. (2012) . Samples were analyzed in a single run, and intra-assay coefficients of variation (CV) were 6.4% for tCa and 4.8% for Mg.

Peripartum Blood Gas Profiles

Check if the same lab product or an alternative is used in the 5 most similar protocols
Whole blood was sampled by puncture of the jugular vein from all cows at 254 d of gestation, the day before switching cows assigned to the short treatments to their respective negative DCAD diets. Whole blood was sampled again from a subset of the first 21 blocks comprising 80 cows (-70_short = 21, -180_short = 20, -70_long = 19, and -180_long = 21) at 268 and 272 d of gestation, corresponding to -10 and -6 d relative to expected calving, and at 0, 1, 2, 3, and 4 d postpartum. Samples were analyzed within 1 to 2 min for concentrations of iCa, Na, K, pH, bicarbonate, base excess, total dissolved CO 2 (tCO 2 ), saturation of O 2 (sO 2 ), and partial pressures of O 2 and CO 2 (pCO 2 ) using a hand-held biochemical analyzer (VetScan i-STAT, Abaxis, Union City, CA).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
