Phosphate-Buffered Saline Preparation and Polymer Characterization

Phosphate-buffered saline
(PBS) solutions were prepared using pre-formulated tablets (Sigma-Aldrich)
in 200 mL of Milli-Q water (>18.2 Ω mean resistivity) to
give [NaCl] = 0.138 M, [KCl] = 0.0027 M, and pH 7.4. Poly(methyl vinyl
ether-alt-maleic anhydride) Mn 80 000 and 311 000 g.mol–1, dimethylaminoethanol, and N-[4-(aminomethyl)benzyl]rhodamine
6G-amide bis(trifluoroacetate) were purchased from Sigma-Aldrich.
Poly(methyl vinyl ether-alt-maleic anhydride) average Mn 20 000 g mol–1 was
purchased from Scientific Polymer Products; the dispersity of polymer P2 from the manufacturer is 2.7 and that of P3 is 3.47; the supplier of P1 did not list the Mw value, only the Mn. Sodium chloride was purchased from Fisher Scientific. All solvents
were purchased from VWR or Sigma-Aldrich, and the reagents were used
without further purification unless indicated.

Cytotoxicity Evaluation of Polymeric Compounds

Advanced Dulbecco’s
Modified Eagle’s
Medium, Ham’s F-12K media, and penicillin/streptomycin/amphotericin
B were obtained from Gibco. Poly(ethylene glycol) (Mn 8 kDa) was obtained from MP Biomedicals. Dimethylaminoethanol,
dimethyl sulfoxide, fetal bovine serum, Dulbecco’s phosphate-buffered
saline, poly(ethylene glycol)methyl ether (Mn 20 kDa), and poly(methyl vinyl ether-alt-maleic anhydride) (Mn 80 kDa) were obtained
from Sigma-Aldrich. A live/dead viability/cytotoxicity kit and CellEvent
Caspase-3/7 Green Detection Reagent were obtained from Thermo Fisher.
All solvents were purchased from VWR or Sigma-Aldrich, and reagents
were used without further purification unless indicated.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
