Plasma IL-27 Quantification by ELISA

Check if the same lab product or an alternative is used in the 5 most similar protocols
Plasma IL-27 level was measured with pre-coated LEGEND MAX Human IL-27 ELISA Kit (BioLegend Inc., San Diego, CA, USA) in accordance with the manufacturer's instructions. Standard curve was generated by using serial diluted standards. Each plate was run on an independent standard curve for reference. Plasma IL-27 concentration was calculated by generated standard curve. The minimum detectable level of the ELISA kit was 11 pg/ml and the kit had no cross-reaction with 14 other human cytokines [25] .

Plasma IL-27 and hsCRP Quantification

Check if the same lab product or an alternative is used in the 5 most similar protocols
Blood samples were collected in EDTA-containing tubes, and the plasma was stored at –80°C. Plasma IL-27 levels were measured by an enzyme-linked immunosorbent assay (ELISA) using a commercially available kit (LEGEND MAX Human IL-27 ELISA Kit; BioLegend, San Diego, CA) at Ochanomizu University in accordance with the manufacturer's instructions. The minimum level detected by this assay was 0.01 ng/mL. The intra-assay and inter-assay coefficients of variation were <6.0% and <5.5%, respectively. Plasma hsCRP levels were measured by a BNII nephelometer (Dade Behring, Tokyo, Japan).

Quantification of Inflammatory Cytokines in MDM Cultures

Check if the same lab product or an alternative is used in the 5 most similar protocols
The ELISA MAX™ Deluxe Set for Human TNFα,IL6, MCP-1/CCL2 and CCL5 (RANTES) (BioLegend, San Diego, CA, United States) was used for detection of TNFα,IL6, MCP-1/CCL2 and CCL5 proteins in supernatants of MDMs cultures, following manufacturer´s instructions. The detection limit was 2–4 pg/ml. The LEGEND MAX™ Human IL27 ELISA Kit (BioLegend, San Diego, CA, United States) was used for the detection of mature IL27 protein in supernatants of MDMs cultures, following the manufacturer’s instructions. The detection limit was 11 pg/ml.

Quantification of IL-27 and IL-23 in Serum

Check if the same lab product or an alternative is used in the 5 most similar protocols
The serum of blood samples was collected from each subject, and the concentration of IL-27 was quantified by using a LEGEND MAX™ Human IL-27 ELISA kit (BioLegend, San Diego, CA). The concentration of IL-23 was detected by using a LEGEND MAX™ Human IL-23 ELISA kit (BioLegend, San Diego, CA). The protocol was performed according to the manufacturer's protocols. The sensitivity of the ELISA kits was 11 pg/ml for IL-27 and 2.45 pg/ml for IL-23.

Quantifying IL-27 and CRP Biomarkers

Check if the same lab product or an alternative is used in the 5 most similar protocols
Blood samples were collected into tubes with EDTA and centrifuged at 2000× g for 15 min at 4 °C. Plasma was frozen and stored at −80 °C until use. For the measurement of IL-27 levels, the enzyme-linked immunosorbent assay (ELISA) (LEGEND MAX™ Human IL-27 ELISA Kit; BioLegend, San Diego, CA, USA) was used. As previously reported [21 (link)], we assessed IL-27 levels at Ochanomizu University according to the manufacturer’s instructions. According to data from the manufacturer, the lowest detection limit of this kit was 0.01 ng/mL. The intra- and interassay coefficients of variation were found to be <6.0% and <5.5%. To measure high-sensitivity C-reactive protein (CRP) levels, a BNII nephelometer (Siemens Healthineers, Tokyo, Japan) was used.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
