Immunofluorescence Staining of AhR in Cardiomyocytes

Check if the same lab product or an alternative is used in the 5 most similar protocols
Cardiomyocytes were cultured onto a 24-well plate with glass cover slips at a density of 5 × 104 cells/well. The cells were then fixed with 4% paraformaldehyde for 20 min and quenched with 1 M ethanolamine diluted in PBSc. After washing, cells were blocked with 0.5% bovine serum albumin in PBS for 30 min, then the blocking solution was removed and the cells were incubated overnight at 4 °C with rabbit anti-AhR (1:100 dilutions, BioWorld). Cells were washed and incubated with mouse anti-rabbit IgG-TR (1:1000 dilutions, Santa Cruz Biotechnology) at room temperature for 1 h. Then, the cells were gently washed under the cover slip three times with PBS and visualized under a laser scanning confocal microscope (Fluoview FV1000 confocal system, Olympus).

Immunofluorescence Staining of ER-alpha and NF-κB

Check if the same lab product or an alternative is used in the 5 most similar protocols
Cells cultured on cover slides were fixed with 2% paraformaldehyde for 20 minutes, followed by three washes with PBS with Tween (PBST). The cells were then blocked using PBS containing 10% serum and Triton X-100 for 1 hour at room temperature. Subsequently, the cells were incubated with rabbit anti-ER alpha antibody and NF-κB overnight at 4 °C. After washing three times with PBST, the cells were incubated for 45 minutes at room temperature with secondary antibodies that were diluted in the blocking buffer. These secondary antibodies included FITC-mouse anti-rabbit immunoglobulin G (IgG) and mouse anti-rabbit IgG-TR (Santa Cruz Biotechnology). The cells

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
