Quantitative RT-PCR Analysis of Gene Expression

Check if the same lab product or an alternative is used in the 5 most similar protocols
Total RNA was extracted from lung tissue samples or cells with TRIzol. Prime ScriptTM Trimester Mix (Takara, Japan) was utilized for reverse transcription of RNA. SYBR Premix Ex TaqTM11 (Takara, Japan) and a Step One Plus RT-PCR system (Life Technologies, USA) were utilized to perform RT-PCR based on the manufacturer’s instructions. The results were normalized to those for β-actin, and quantification was performed with the 2-ΔΔCt of each sample/2-ΔΔCt of Ctrl method. The primers are shown in Table S3.

Quantitative Analysis of TGF-β1 Expression in MSCs

Check if the same lab product or an alternative is used in the 5 most similar protocols
Total RNA from MSCs, MSC-NC, and MSC-TGF-β1 was extracted using TRIzol reagent (Life Technologies, USA). RNA was reverse transcribed using Prime Script TM Trimester Mix (Takara, Japan) and subjected to RT-PCR with SYBR Premix Ex TaqTM11 (Takara, Japan) and a Step One Plus RT-PCR system (Life Technologies, USA). β-Actin was used as an endogenous control. RT-PCR primers were synthesized by Sangon Biotech (Shanghai, China) and the sequences were as follows: mouse β-actin forward primer, 5′-GGGAAATCGTGCGTGAC-3′, and reverse primer, 5′-AGGCTGGAAAAGAGCCT-3′, and mouse TGF-β1 forward primer, 5′-AGACGGAATACAGGGCTTTCGATTCA-3′, and reverse primer, 5′-CTTGGGCTTGCGACCCACGTAGTA-3′.

Analyzing Gene Expression in RAW264.7 Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
Total RNA from RAW264.7 cells was extracted using TRIzol reagent (Life Technologies, USA). Reverse transcription of RNA and RT-PCR was performed by using Prime Script TM Trimester Mix (Takara, Japan) and SYBR Premix Ex TaqTM11 (Takara, Japan) with a Step One Plus RT-PCR system (Life Technologies, USA) according to the manufacturer’s instructions. β-actin was used as an endogenous control. Primers for RT-PCR were synthesized by Sangon Biotech (Shanghai, China). The primer sequences are as follows: mouse b-actin forward primer, 5′-GGGAAATCGTGCGTGAC-3′ and
reverse primer, 5′-AGGCTGGAAAAGAGCCT-3′;
mouse ARG-1 forward primer, 5′-AACACTCCCCTGACAACCA-3′ and

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
