VEGFR2 Silencing in HUVEC Proliferation

Check if the same lab product or an alternative is used in the 5 most similar protocols
Silencer Select small interfering RNAs (siRNAs) against VEGFR2 (s7823; Thermo Fisher Scientific) and Silencer Select negative-control siRNA number 2 (Thermo Fisher Scientific) were used for RNAi assays. Briefly, 5,000 cells of HUVEC in EGM-2 were plated in each well of a 96-well plate and incubated at 37°C for 6 h. The medium was replaced with 0.1 mL of M199–10% FBS, and the cells were transfected with siRNA (0.5 pmol/well) using Lipofectamine RNAiMAX (Thermo Fisher Scientific) in accordance with the manufacturer's instructions. After 24 h, the cells were washed and replaced with fresh M199–10% FBS and then used for cell proliferation assays against VEGF-A165 and BafABba. VEGFR2 knockdown at 24 h after transfection was confirmed by quantitative reverse transcription-PCR (qRT-PCR).

siRNA Transfection in HuH-7 Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
Unless mentioned otherwise, HuH-7 cells were transfected with 10 nmol/L Silencer Select Small interfering RNAs (siR-NAs) (ThermoFisher) or nontarget control using Lipofectamine RNAiMAX Reagent for 72 hours according to the manufacturer's instructions. After harvest, the RNA and protein were isolated.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
