Volatile Profiling of Chinese Rose Cultivars

The Chinese rose cultivars have different colors and fragrances, and the four cultivars selected in this study had large color differences. Four cultivars (Rosa ‘Funkuhr’, R. ‘Pink Panther’, R. ‘Crimson Glory’, and R. ‘Blue River’, denoted RF, RPP, RCG and RBR, respectively) were generously provided by the experimental field of Guizhou Botanical Garden and harvested at E 106° 40’–27”; N 26° 11’ 39”, located in Guiyang City, China (Figure 1). The flowers were harvested before petal opening and transferred to the laboratory. The samples were powdered following the method described in literatures (Qian et al., 2019 (link); Gil et al., 2020 (link)). Methanol was purchased from Sigma−Aldrich Fluka (Buchs, Switzerland). Methyl hexadecanoate (internal standard; purity ≥ 97.5%) was obtained from CATO (Portland, OR, USA) and stored at 4°C until use. Commercially available SPME coating fibers, including 85 µm carboxen/polydimethylsiloxane (CAR/PDMS), 50/30 µm divinylbenzene/carboxen/polydimethylsiloxane (DVB/CAR/PDMS), 65 µm polydimethylsiloxane/divinylbenzene (PDMS/DVB), and 60 µm carbowax-polyethylene glycol (PEG), were purchased from Supelco (Bellefonte, PA, USA).

Comprehensive Analysis of Acorus villosum Samples from Yangchun, China

Yangchun is located in southwest of Guangdong province of China (111° 16′ 27″ to 112° 09′ 22″ E, 21° 50′ 36″ to 22° 41′ 01″ N), which is the geo-authentic habitats of A. villosum. Seventy-two batches of fresh A. villosum samples containing fruits (1–18), roots (19–36), leaves (37–54) and stems (55–72) were obtained from different places of Yangchun. The detailed characteristics are presented in Table 1. Species identification was performed by Dr. Jing Zhao and Dr. Hao Hu. The voucher specimens of these samples were deposited at the Institute of Chinese Medical Sciences, University of Macau, Macao SAR, China.

All other chemicals and reagents were of analytical grade. The extraction fibers: 100 μm polydimethylsiloxane (PDMS), 65 μm polydimethylsiloxane/divinylbenzene (PDMS/DVB), 85 μm polyacrylate and 75 μm carboxen poly(dimethylsiloxane) (CAR/PDMS) were purchased from Supelco (Bellefonte, PA, USA). Headspace vials (20 mL) and accessories were obtained from Agilent (Palo Alto, CA, USA).

Volatile Profiling of Zhejiang and Guizhou SD

A total of 11 batches of Zhejiang SD (batch number ZJ01, ZJ02, ZJ03, ZJ04, ZJ05, ZJ06, ZJ07, ZJ08, ZJ09, ZJ10, and ZJ11) and 13 batches of Guizhou SD (batch number GZ01, GZ02, GZ03, GZ04, GZ05, GZ06, GZ07, GZ08, GZ09, GZ10, GZ11, GZ12, and GZ13) were purchased from Hangzhou Chinese Herbal Medicine Trading Center (Zhejiang Province) and Guiyang Chinese Herbal Medicine Market (Guizhou Province). These samples were authenticated by Zhang Yujin (an associate professor from Zunyi Medical University) and deposited in the chemical laboratory. n-Hexane of analytical grade was purchased from Suzhou Boyuan Chemical Co., LTD (Suzhou, China). n-Alkane (C8–C30) series were provided by Sa Lan Chemical Co., LTD (Shanghai, China). SPME holder and 100 µm Polydimethylsiloxane (PDMS), 75 µm Carboxen/Polydimethylsiloxane (CAR/PDMS), 65 µm Polydimethylsiloxane/Divinylbenzene PDMS/DVB, and 50/30 µm Divinylbenzene/Carboxen/ Polydimethylsiloxane (DVB/CAR/PDMS) fiber were purchased from Supelco Inc (Bellefonte, PA, USA). HS-SPME unit and transparent sample bottle were purchased from Kanglin Technology Co., LTD (Beijing, China).

Volatile Organic Compounds Analysis Protocol

The amines TMA, DMA, and the internal standard, n-propylamine hydrochlorides, were acquired from BDH and Merck. Sodium chloride (NaCl, 99.5%) and sodium hydroxide (NaOH) were obtained from PanReac (Barcelona, Spain). Ultra-pure water was obtained from a Milli-Q® system (Millipore, Burlington, MA, USA), and helium of purity 5.0 was used as the GC carrier gas, supplied by Air Liquide (Alges, Portugal). The SPME holder for the manual sampling of SPME fiber and the respective fibers, namely the divinylbenzene/carboxen/polydimethylsiloxane (DVB/CAR/PDMS), with a 50/30 mm film thickness, carboxen/polydimethylsiloxane (CAR/PDMS) with a 75 mm film thickness, and polydimethylsiloxane/divinylbenzene (PDMS/DVB) with a 65 mm film thickness, were purchased from Supelco (Bellefonte, PA, USA). Ringer solution (Oxoid, Basingstoke, Hampshire, UK) was used for microorganism transference. Inoculations were made in iron agar, nutrient agar (Oxoid, Basingstoke, Hampshire, UK), and MacConkey agar (PanReac AppliChem, Darmstadt, Germany). For physical evaluation, a Torrymeter 295 (Distell, West Lothian, Scotland, UK) was used.

Simultaneous Analysis of Fragrance Allergens, Synthetic Musks, Preservatives, and Plasticizers

The 60 target compounds (23 fragrance allergens, 11 synthetic musks, 10 preservatives and 16 plasticizers), their CAS number, the retention time, the molecular mass and the MS/MS transitions are depicted in Table S1. Methanol, ultrapure water MS grade and ethyl acetate were supplied by Scharlab (Barcelona, Spain) and acetone by Sigma Aldrich Chemie GmbH (Steinheim, Germany). Individual stock solutions were prepared in methanol and further dilutions and mixtures in acetone (spike solutions). All solutions were stored in amber glass vials and protected from light at −20 °C. All solvents and reagents were of analytical grade.
Commercial 65 μm polydimethylsiloxane/divinylbenzene (PDMS/DVB), 50/30 μm divinylbenzene/carboxen/polydimethylsiloxane (DVB/CAR/PDMS) and 85 μm polyacrylate (PA) fibres and manual SPME holders were obtained from Supelco (Bellefonte, PA, USA). Prior the first use, the fibres were conditioned as recommended by the manufacturer, inserting them in the GC injector under helium flow at 250 °C (PDMS/DVB), 270 °C (DVB/CAR/PDMS) and 280 °C (PA) for 30 min.
