Immunoblotting Analysis of H+,K+-ATPase Subunits

Check if the same lab product or an alternative is used in the 5 most similar protocols
H+, K+-ATPase subunits detection was performed as described previously [15 (link)]. For signaling pathway analysis, PANC-1 cell lysate was boiled 3 min at 95 °C. Primary antibodies against gastric HKα1 (Abcam EPR12251, 1:1000), non-gastric HKα2 (C384-M79, 1:1000, kindly donated by J. J. H. H. M. De Pont and H. G. P. Swarts) [56 (link)], HKβ (Sigma A274, RRID:AB_258027, 1:1000), β-Actin (Santa Cruz Sc-47778, 1:1000, RRID: AB_626632), pSTAT3 (Cell Signaling #9145, 1:1000, RRID: AB_2491009), STAT3 (Cell Signaling #9139, 1:1000, RRID: AB_331757) and Cyclin D1 (Millipore #06-137, 1:500, RRID: AB_2070695) were used.

Mature Human Neuron Differentiation from iPSCs

Check if the same lab product or an alternative is used in the 5 most similar protocols
To obtain mature human matured neurons in a culture system, ReproNeuro, a neuron progenitor derived from human iPSCs, was purchased from ReproCELL (Yokohama, Japan) and maintained in ReproNeuro maturation medium for 14 days according to the manufacturer's instructions. All assays were performed on cells between passages 3 and 12 and were repeated at least 3 times in duplicate or triplicate. LPA (18:1) was purchased from Cayman Chemical (Ann Arbor, MI); LPA was dissolved in phosphate-buffered saline containing 0.1% lipid-free bovine serum albumin (Sigma-Aldrich, MO) to generate a 10-mM stock solution. GW9662 and Ki16425 were purchased from Sigma-Aldrich (St. Louis, MO).
We used antibodies against PPARγ (rabbit monoclonal, sc-7196) and KLF9 (rabbit monoclonal, GTX129316), horseradish peroxidase (HRP)-conjugated anti-mouse and antirabbit secondary antibodies (Cell Signaling Technologies, Danvers, MA), and an antibody against β-actin (mouse monoclonal, sc-47778).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
