Biochemical Analysis of Rat Serum

Check if the same lab product or an alternative is used in the 5 most similar protocols
After TFN treatment for four weeks, rats in all groups were anesthetized with 10% chloral hydrate (4.0 ml/kg) on the 29th day. Blood samples were collected from the aorta abdominalis and transferred into the tube with heparin sodium. Blood samples were subsequently centrifuged at 3,500 rpm for 15 min at 4°C and the upper serum was collected for IL-1β, TNF-α, SOD, MDA, Scr, and BUN analysis. Olympus AU640 automatic biochemical analyzer was applied to disclose Scr and BUN levels. The blood of IL-1β, TNF-α, SOD, and MDA content was detected by ELISA according to the instruction in the kits.

Uric Acid Quantification via Uricase

Check if the same lab product or an alternative is used in the 5 most similar protocols
The uricase method was used to detect uric acid levels. We got the proband's blood under fasting. The blood was centrifuged at 3000 r/min for 10 min. Then the supernatant was taken and detected by an Olympus AU640 automatic biochemical analyzer. The kit was purchased from Mindray (Shenzhen, China).

High-Fat Diet Induces Obesity

Check if the same lab product or an alternative is used in the 5 most similar protocols
After adaptive feeding for one week, the C57bl/6j mice were fed with high fat forage (fat content about 36%) for 8 weeks up to the obesity model standard. The mice weight was gained at the 4th and 8th week, respectively. The bloods in each group were collected at the 4th and 8th weekend to measure the serum uric acid levels by Olympus AU 640 automatic biochemical analyzer.

Homocysteine Determination in Patients

Check if the same lab product or an alternative is used in the 5 most similar protocols
The enzymatic cycling assay was used to determine the level of homocysteine in the patient (Roberts & Roberts, 2004). The patient serum was obtained as described in 2.5.2 and detected by an Olympus AU640 automatic biochemical analyzer. The homocysteine detection kit was purchased from Medicalsystem (Zhejiang, China).

Thyroid and Metabolic Biomarker Analysis

Check if the same lab product or an alternative is used in the 5 most similar protocols
The clinical characteristics of the study population include in serum free triiodothyronine (FT3), serum free thyroxine (FT4), alanine aminotransferase (ALT), aspartate aminotransferase (AST), triglyceride content (TG) and total cholesterol (TC). These were measured by Olympus AU640 automatic biochemical analyzer (Olympus Corporation, Tokyo, Japan).

Liver Enzyme and Lipid Analysis

Check if the same lab product or an alternative is used in the 5 most similar protocols
Liver tissue (200 mg) was homogenized in 2 ml 0.7% saline and centrifuged at 3,000 × g for 15 min at 4°C. The protein-containing supernatant was used to measure enzyme activities (AMPK, ACC, FAS, CPT-1, and ACO) using customized ELISA kits (Zike Biological Technology Co., Ltd., Shenzhen, China) according to the manufacturer’s instructions.
The concentration of plasma triglycerides (TG), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), and total cholesterol (TC) was measured within 24 h of blood collection in a refrigerator by an Olympus AU640 automatic biochemical analyzer (Olympus, Tokyo, Japan).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
