Methadone Hydrochloride Purification Protocol

Methadone hydrochloride was purchased from Esteve Pharmaceuticals S.A. (Barcelona, Spain). Methylparaben and propylparaben, acquired from Fagron Iberica (Terrassa, Spain), were used as preservatives. Purified water was obtained from Grifols laboratory (Barcelona, Spain). All were of Pharmacopoeia grade. In the mobile phase, we used acetonitrile HPLC gradient grade purchased from VWR Prolabo Chemicals (Fontenay-Sous-Bois, France). Phosphoric acid, sodium hydroxide (>99%), hydrochloric acid, and hydrogen peroxide were supplied from Panreac (Barcelona, Spain) and Milli-Q water. All reagents and solvents were of analytical grade.

HPLC Analysis of Carbohydrates and Organic Acids

Glucose, fructose, ethanol, glycerol and acetic acid were individually analysed, using a Varian high performance liquid chromatography (HPLC) system, equipped with a Rheodyne injector with a 20 µL loop, a Supelco Gel C—610 H column (300 mm × 7.8 mm) maintained at 35 °C and a refractive index detector RI-4 (Varian, Palo Alto, CA, USA). Isocratic elution was employed with a mobile phase consisting of 0.1% (v/v) phosphoric acid (Panreac, Barcelona, Spain) at a flow rate of 0.5 mL·min−1. Data were recorded and integrated using Star Chromatography Workstation software (Varian). Glucose, fructose, ethanol, glycerol and acetic acid were quantified by external standard calibration.

Comparative Evaluation of Ibuprofen Formulations

Four commercially available immediate release ibuprofen-containing products were investigated. The formulations were purchased from pharmacies in the United States and Hungary. The tested products and their active ingredients are listed in Table 1.
Ibuprofen drug substance was purchased from Sigma–Aldrich (Burlington, VT, USA). All chemicals used were of analytical grade. The following chemicals were used: sodium-hydroxide; sodium-chloride; sodium dihydrogen phosphate monohydrate; hydrochloric acid; (Molar Chemicals Kft., Budapest, Hungary); acetonitrile (PanReac AppliChem, Darmstadt, Germany); phosphoric acid; (Emsure ACS. Reag. Ph. Eur., Budapest, Hungary); SIF powder (BiorelevantTM, London, UK); and Pepsin (Sigma–Aldrich, Burlington, VT, USA).

HPLC-based Quantification of Pharmaceutical Compounds

Amiloride hydrochloride and (±)-propranolol hydrochloride were purchased from Sigma-Aldrich (St. Louis, MI, USA). HPLC-grade methanol used for the preparation of stock and working solutions and the HPLC grade acetonitrile used for the mobile phase was provided by Romil (Cambridge, England). For the preparation of the extraction buffer solution, potassium dihydrogen phosphate (>99%) obtained from Merck (Darmstad, Germany) and phosphoric acid (85%) supplied by Panreac (Barcelona, Spain) were used. Ultrapure analytical water was obtained from a Milli-Q Element A10 system (Millipore, Milford, CT, USA). α-cyano-4-hydroxycinnamic acid (CHCA) and DAN (1,5-Diaminonaphthalene) from Sigma-Aldrich and DHB (2,5-Dihydroxybenzoic acid) supplied by Acros Organics (Geel, Belgium) were used as ionizable matrices for MALDI-IMS experiments. Standard stock solutions of 1000 mg/L were prepared in methanol for each analyte separately. With those solutions, 50 mg/L working solutions in methanol were prepared.

Hoechst 33342, Alexa Fluor 594 Phalloidin, LDH and alamar blue assay kits were purchased from Thermo Fisher Scientific (Waltham, MA, USA). Vero (ATCC® CCL-81), HaCat (ATCC® CCL-2404) and HFF-1 (ATCC® SCRC-1041) cells were obtained from ATCC (St. Cloud, MN, USA).
