Picrosirius Red and Immunofluorescence Staining

Western Blot Analysis of DNA Damage Response Proteins

Cells were lysed in RIPA buffer containing 1% PMSF and phosphatase inhibitor cocktail buffer (Beyotime, China). Proteins were loaded onto SDS-PAGE gel for electrophoresis and then transferred onto a PVDF membrane. The blots were probed with primary antibodies at 4 °C overnight followed by incubation with secondary antibodies at room temperature for 1 h. The signal was visualized using enhanced chemiluminescence (Invitrogen, Carlsbad, CA, USA) and quantified by densitometry using Image Lab software.
