Western Blot Analysis of Adipogenic Proteins

Total protein was extracted using RIPA buffer and separated on a 10% polyacrylamide gel, followed by transfer to a PVDF membrane. The primary antibodies used were rabbit anti-PPARγ (Invitrogen, Carlsbad, CA, USA), anti-C/EBPα (Novus Biologicals, Centennial, CO, USA), anti-UCP1 (Abcam, Cambridge, UK), and anti-β-actin (Novus Biologicals, Centennial, CO, USA). HRP-conjugated antimouse IgG or anti-rabbit IgG (Bio-Rad) were used as the secondary antibodies. The blots were treated with ECL substrate solutions and exposed to a ChemiDoc XRS System (Bio-Rad) to detect chemiluminescence.
