Protein expression in serum samples was determined by ELISA. The serum levels of TNF, IL6, IFNγ, IL1β, ICAM1, IL17A, MMP9, IL4, IL12, TGFβ (Thermo Fisher Scientific, US), and HMGB1 (Arigo Biolaboratories, China) were determined by ELISA following the manufacturer instructions. The statistical analyses were carried out using GraphPad Prism 8.0. All experiments were performed by a two-tailed Student’s t-test. P<0.05 was considered significant.

Mixed-sex cohorts of 8–10-week-old Gsdmd−/− and wild-type littermate mice were treated with intraperitoneal LPS from E. coli serotype O111:B4 at 20 mg ml−1 or saline vehicle. Both wild-type and knockout animals were treated with the same preparation of LPS or vehicle control. All animals survived to 6 h post-treatment at which point they were humanely euthanized for plasma and tissue collection. All animals were included in the analyses, with the investigators blinded to both genotype and treatment groups.
